Objective The study aimed to explore the mechanism of IL-1? induced by FGF-7 in Ha CaT;The role of FGF-7 specific antibody(F-9)in skin inflammation was studied in IMQinduced psoriasis-like mouse model.Methods(1)IHC was used to test the changes of FGF-7 and p-FGFR2 level in healthy and psoriasis skin;Immunofluorescence assay was used to detect the colocalization of IL-1? and CD11C/CD163/K14 in psoriasis skin lesions;(2)WB was used to detect the inhibitory effect of FGF-7 specific neutralizing antibody F-9 on FGFR phosphorylation in Ha Ca T.(3)QPCR was used to detect m RNA expression of IL-1? and its receptor antagonist IL-1Ra,WB was used to explore the mechanism of IL-1? induced by FGF-7 in Ha Ca T.(4)psoriasiform mouse model was established by IMQ application on the backs and ears of balb/c mice,HE,IHC,WB,QPCR was used to comprehensively evaluate the therapeutic effect of F-9 on inflammatory mice.Result(1)The expression of FGF-7 and p-FGFR2 in psoriatic skins is significantly upregulated compared with heathy skins,IL-1? is mainly produced by keratinocytes in psoriatic skins.(2)4 ?g/ml F-9 completely inhibited FGF-7-induced phosphorylation of FGFR2.(3)FGF-7 induced IL-1? m RNA levels upregulated in Ha Ca T cells,but downregulated IL-1Ra expression;FGF-7 affects IL-1? expression via FGF-7/FGFR2-PI3K/AKT signaling pathway.(4)FGF-7 antibody F-9 significantly extenuated skin thickening,redness in IMQinduced psoriasiform inflammation mouse model;F-9 also down regulated the expressions of FGFR2,IL-17 and IL-1?.in model mice.
