Research background:(1)Scientific development is inseparable from scientific research activities,and Chinese medicine is no exception.Looking at the development history of traditional Chinese medicine,whenever academic theories develop rapidly,medical scientific research must play an important role.Since the 1950 s,Chinese medicine has attached great importance to the application of modern science and technology and modern pharmaceutical methods in the research of traditional Chinese medicine.It follows the basic principles of random,controlled,double-blind,and repetitive scientific research,and uses statistical processing methods to make research The results are more accurate and reliable.Dianthus superbus is a Dianthus plant in the family Dianthus.Its Latin name Dianthussuperbus L.It is also the dry aerial part of Dianthus D.chinensis L.Dianthus superbus is mainly distributed in Liaoning and Hebei.Its harvesting season is summer and autumn,and it is non-toxic.Dianthus superbus' s main medicine is the upper part of the dry land.Dianthus superbus belongs to cold medicine and has a bitter taste;it belongs to the Heart Meridian and the Small Intestine Meridian.Dianthus superbus has the effects of phlegm diuresis,catarrhal bleeding,etc.At present Dianthus superbus is mainly adapted to clinical features such as blood leaching,menstrual stasis,heat leaching,unpleasant urination,stone leaching,leaching and astringent pain.As a traditional Chinese medicinal material,Dianthus superbus is used in clinical practice,and it has great significance to further clarify the effective chemical components of the medicinal material.(2)The study of Dianthus superbus Chinese herbal medicines comes from the quality control system of Paishi granules of traditional Chinese medicine varieties in Jiangxi Province's key research and development plan projects(No.20171BBG70104).Dianthus superbus is one of the ingredients of Paishi granules,and also provides quality control systems for Paishi granules.Further theoretical data support was made in order to establish better quality control standards for row stone particles.Research purpose: According to the research on the chemical composition of Dianthus superbus in recent years,Dianthus superbus mainly contains volatile oil,triterpenoid saponin,flavonoids and cyclic peptides.These types of compounds play a very important role in Dianthus superbus' s pharmacologically active ingredients.However,Dianthus superbus' s other medicinal ingredients are not yet clear.Based on previous research,this paper uses scientific thinking to better excavates the medicinal activity of Dianthus superbus and explores other medicinal chemical ingredients contained in Dianthus superbus.Research methods: By consulting relevant references,we find that the extraction method of Dianthus superbus is mainly steam distillation,methanol extraction,petroleum ether extraction and other methods.Based on previous research,we mainly use dichloromethane to extract,isolate,and extract Dianthus superbus.We extract the dried roots and leaves of Dianthus superbus by refluxing with 75% ethanol(10 times the amount),combine the extracts,concentrate to obtain a dry extract(extract)and weigh.Take the dry extract,add water,extract 3 times with dichloromethane,recover the solvent,and finally obtain the Dianthus superbus dichloromethane extract.The dichloromethane extract was mixed with silica gel and passed through a silica gel column;after washing with petroleum ether: ethyl acetate in this order,the related compounds were isolated and purified.Then,the obtained compounds and their properties were studied.The spectroscopy method was used in combination with electrospray mass spectrometry and nuclear magnetic resonance spectroscopy to identify the structure and chemical properties of the isolated compounds.Research results: A total of 14 monomer compounds were obtained,and these compounds were identified as: We identified 14 chemical agents,including:(+)-epipinoresinol(1)?pinoresinol(2)?dibutyl phthalate(3)?(3Z,6Z,9Z)-3,6,9-nonadecatriene(4)?catechol(5)?dehydrololiolide(6)?aurantiamide acetate(7)?quillaic acid(8)?dehydrovomifoliol(9)?succinic acid(10)?melrubiellin A(11)?melrubiellin B(12)?salicylic acid(13)?Prinsepiol(14).Except catechol and salicylic acid,others were isolated from Dianthus superbus for the first time.By studying the chemical properties of the extracted compounds,they can better excavate their medicinal value.Research conclusion: In this thesis,new monomer compounds found in Dianthus superbus were studied,and its chemical properties and current application scope were described in detail.The pharmacological mechanism of active ingredients contained in Dianthus superbus was clarified for future researchers.It provides a reference basis for clinical effects,and provides scientific data support for the research on the quality control system of Pai Shi particles.
