This paper was composed by two parts.The first part was about the active compounds study of anti-rheumatoid arthritis(RA)effect of the rhizomes of Impatiens pritzellii Hook.f.var.hupehensis Hook.f.The second part was about the antineoplastic active compound of Dianthus superbus L.(Part one)The rhizomes of Impatiens pritzellii Hook.f.var.hupehensis Hook.f.(Balsaminaceae)was a popular folk medicine with the effect of dehumidify and dispel the wind,desslpate blood stasis and swelling in Tujia-tribe district of Hubei Province as an anti-RA medicine,the curative effect is really good.We achieved the right results,after systematically studied the chemical constituents and the medicinal effect of this plant based on previous studies.Through chromatography separation and spectral identification methods,Our chemical constituents study of I.pritzellii afforded 14 compounds from the active fraction of anti-RA(n-BuOH extracts):2,6-dimethyl-2-vinyl-2,3,4,7-tetrahydrooxepine(1),1,3,6-trihydroxy-7-methyl-anthraquinone(2),4-hydroxybenzaldehyde(3),4-(3-methoxy-4-hydroxyphenyl)-2-butanone(4),Podophyllotoxin(5),Scopoletin(6),?-spinasterol(7),3-O-?-D-glucopyranosyl-?-spinasterol(8),3-O-[(6-methoxy)-?-D-glucuronopyranosyl] echinocystic acid(9),3-O-?-D-glucuronopyranosyl-echinocystic acid 28-O-?-D-xylopyranosyl-(1?4)-[3???-acetoxy]-?-L-rhamnopyranosyl-(1?2)-?-L-arabinopyranoside(10),3-O-[(6?-methoxy)]-?-D-glucuronopyranosyl ester-echinocystic acid 28-O-?-D-xylopyranosyl-(1?4)-[(3???-acetoxy)]-?-L-rhamno-pyranosyl-(1?2)-?-L-arabinopyranoside(11),3-O-[(6?-methoxy)]-?-D-glucuronopyranosyl ester-echinocystic acid 28-O-?-D-xylopyranosyl-(1?4)-[(3???-acetoxy)]-?-L-rhamnopyranosyl-(1?2)-?-D-xylopyranoside(12),3-O-?-D-glucuronopyranosyl-echinocystic acid 28-O-?-D-xylopyranosyl(1?4)-?-L-rhamnopyranosyl-(1?2)-?-L-arabinopyranoside(13),3-O-?-D-glucuronopyranosyl-echinocystic acid 28-O-?-D-xylopyranosyl-(1?4)-?-L-rhamnopyranosyl-(1?2)-?-D-xylopyranoside(14).We built a screening model of T lymphocyte proliferation,and studied the 6 saponins obtained from the active fraction of anti-RA effects on inhibiting proliferation of T lymphocytes,all of the six triterpenoid saponins could suppress the proliferation of T lymphocytes,the dose-effect relationship was significant with the concentration between 5?g/ml and 100 ? g/ml,inhibitory effects enhanced with the increasing drug concentrations.The inhibition ratio of six saponins(compound 9,10,11,12,13,14)was 22.26%,32.74%,36.83%,27.93%,37.92% for T lymphocytes proliferation,respectively,all of them showed varying degree of drug efficacy.Fourteen compounds had been isolated and identificated from the active fraction of anti-RA of I.pritzellii.four compounds of them was new structure,eight compounds was obtained from this plant for the first time.The experimental result of inhibiting proliferation of T lymphocytes indicated that 6 saponins showed good drug efficacy,and 5 saponins of them were discovered inhibiting proliferation of T lymphocytes for the first time.It possessed important significance for annotating therapeutic material basis of anti-RA effect of I.pritzellii.,and providing a experimental foundation for exploitation of the ethnomedicine.(Part two)Dianthus superbus L.(Caryophyllaceae)which is commonly used as a traditional Chinese medicine for a diuretic and treating stranguria,activate blood to promote menstruation agent.It was listed in all editions of “Chinese Pharmacopoeia” the first volume.According to the modern pharmacology research,dianthi herba has significant antitumor effect.In this paper,on the basis of previous researches,we studied the active antitumor constituents of dianthi herba,obtained good experimental results.Through the interdisciplinary methods combining with Chemistry of Traditional Chinese Medicine,Molecular biology and Analytical Che mistry,under the guidance of antitumor activity screening,tracked the a ntitumor active chemical composition of dianthi herba and researched it.Using solvent extraction method,dianthi herba extract were separ ated into some diffirent fractions,and screened out of the active antitu mor material fractions;Using chromatographic separation method,the key active antitumor material fraction were separated into eight subfra ctions,screened out of some active antitumor material subfraction aga in;and the key active antitumor material subfraction were purified to obtain the monomer,it was identified as asiatic acid(2?,3?,23?-trihydr oxyurs-12-en-28-oic acid)by spectrum technique;and asiatic acid show ed significant inhibition effect of Human Hepatocellular Carcinoma B EL-7402 Cells by MTT assay.Through our research,asiatic acid was discovered as significant an titumor active compound from dianthi herba for the first time,it is im portant for interpretation of the material basis of this traditional chine se medicine,it was expected to supply target compound for antitumor chemical medicine research and development.
