Objective(1)To investigate the relationship between monocyte chemoattractant protein-1(MCP-1),speckled protein 110(SP110)and estrogen related receptor bata(ESRRB),gene single nucleotide polymorphism(SNP)and susceptibility to tuberculosis(TB);(2)To explore the correlation between serum ESRRB expression level and tuberculosis,and the relationship between serum ESRRB expression level and ESRRB gene polymorphism.Methods(1)Using the design of case-control study,we selected the patients who were hospitalized in Beijing Chest Hospital Affiliated to Capital Medical University from February 2018 to April 2019,and A total of 607 new active TB cases were selected “according to the TB diagnostic standard WS 288-2017”,At the same time,660 healthy people(all Han nationality)with a history of close contact with tuberculosis in the same period were selected as healthy controls.Using competitive allele specific PCR(KASP)method,the single nucleotide polymorphisms(SNPs)of rs1024611 of MCP-1 gene,rs1135791,rs11556887 and rs9061 of SP110 gene,rs12437118 of gene ESRRB were analyzed.The distribution of SNPs between the case group and the control group was studied by analyzing the genotype frequency,allele frequency and two genetic models of the case group and the control group.(2)Using the design of case-control study,we selected the patients who were hospitalized in Beijing control group was detected by enzyme-linked immunosorbent assay(ELISA),combined with the detected ESRRB genotyping.To explore the relationship between serum ESRRB concentration and pulmonary tuberculosis,and the relationship between serum ESRRB expression and SNPs.Results(1)After analysis and comparison,there was no significant difference in gender and age between the included case group and control group(P> 0.05);the genotype distribution of the five loci SNPs between the two groups was consistent with Hardy-Weinberg equilibrium(HEW).The distribution frequencies of the three genotypes AA,AG and GG in the ESRRB(rs12437118)locus in the tuberculosis group and the control group were 11.7%,38.1%,50.7% and 9.5%,45.7 %%,44.8%;allele frequency A And G in tuberculosis group and control group were 30.7%,32.3% and 69.3%,67.7%,respectively.The distribution of genotype frequency of this locus in tuberculosis group and control group was significant(c2=7.619,P=0.022);There was no significant difference in the distribution of allele frequencies between the two groups.A gene polymorphism analysis of other 4 loci in the MCP-1 and SP110 genes showed that the genotype and allele frequencies were distributed between the case group and the control group,The difference between them was not significant(P>0.05).In the analysis of the selected 5 SNPs genetic models,we found that the dominant model of rs1135791 locus [(CT+CC)vs TT] was significantly correlated with TB susceptibility(OR = 1.558,95% CI:1.247~1.948,P = 0.001),carrying TT genotype can significantly reduce the risk of TB.(2)A total of 147 serum samples from the tuberculosis group were included in this study,including 87 males(59.2%)and 60 females(40.8%),with an average age of(39 ± 11.6)years,and 147 healthy controls.Including 72 males(48.9%)and 75 females(51.1%),the average age was(37 ± 13.2)years old.After statistical analysis,there was no significant difference in age and gender between the case group and the control group(P> 0.05).The serum ESRRB expression of the two groups was measured by ELISA.The mean serum ESRRB concentration of the TB group was 342.8 ± 27.07 pg /ul,and the mean serum ESRRB concentration of the control group was 469 ± 30.46 pg /ul.After statistical analysis,there were differences in serum ESRRB levels between the two groups,and the ESRRB concentration in TB serum was significantly lower than in the control group,the difference was significant(P < 0.05).Serum ESRRB expression levels were different in people with different genotypes at rs12437718 in TB group.Serum ESRRB concentration of AA genotype was higher than AG and GG.Serum ESRRB concentration of GG genotype was the lowest.The difference was not significant among the three genotypes(P > 0.05).Conclusion(1)the polymorphism of ESRRB(rs12437118)affects the susceptibility to TB,and AA genotype is the susceptible factor;(2)The polymorphisms of MCP-1(rs1024611),SP110(rs1135791,rs11556887,rs9061)were not correlated with the incidence of tuberculosis;(3)The dominant model of rs1135791 locus was significantly related to the susceptibility of tuberculosis.Carrying TT genotype could significantly reduce the risk of tuberculosis;(4)The concentration of ESRRB in the serum of TB patients is significantly higher than that of healthy people;(5)There was no significant correlation between ESRRB rs12437118 polymorphism and serum ESRRB concentration in TB group.
