Objective:Modern molecular biology techniques were used to detect TNF-?,VEGF,IL-2factor and PI3 K protein in rats with ovarian endometriosis cyst model to study the therapeutic effect and efficacy mechanism of self-designed anti-tumor prescription on ovarian endometriosis cyst,so as to provide objective basis for further promotion of this prescription.Methods:60 female SD rats were randomly divided into 6 groups: sham operation group,model group,low dose group,medium dose group,high dose group and cassia tuckaing pill group.OEC rat model was established by autologous endometrium transplantation in the other groups,and the general conditions of rats in each group were observed during 4 weeks of continuous administration.The writhing reaction of rats in each group was observed and recorded after the treatment.The growth of heterotopic lesions in each group was observed and the volume was measured.The pathological conditions of ectopic lesions in each group were observed under light microscope after HE staining.Serum levels of TNF-?,VEGF,IL-2 were determined by Elisa.Immunohistochemical detection of each group of PI3 K protein expression in rat OEC group.Results:1.Twisting body reaction: compared with the model group,the twisting body reaction times in the high,medium and low dose groups and the cassia twig tuckahoe pill group were all reduced(P < 0.05).The inhibition rate was higher in the medium dose group than in the low dose group and the high dose group.2.The volume of heterotopic lesions in each group: there was no statisticaldifference in the volume of heterotopic lesions in OEC in each group before medication(P > 0.05).The volume of heterotopic OEC lesions was reduced in the low-dose,medium-dose and high-dose groups as well as in the cassia twig tuckahoe pill group(P < 0.05).3.Contents of serum TNF-?,VEGF,IL-2 factor and PI3 K protein:The contents of TNF-?,VEGF and PI3 K protein in the serum of the model rats were decreased in the low-dose,medium-dose and high-dose groups and in the cassia twig tuckahoe pill group,which were statistically significant compared with the model group(P < 0.05).The contents of TNF-? and VEGF in serum of the groups with medium and high dose and in the cassia twig tuckahoe pill group,which was better than that in the low-dose self-proposed anti-tumor prescription group.The difference was statistically significant(P < 0.05),among them,the content of TNF-? in the medium and high dose groups was lower than that in the cassia twig tuckahoe pill group(P < 0.05),while there was no difference in serum VEGF level between the medium and high dose groups and the cassia twig tuckahoe pill group(P > 0.05).The content of PI3 K protein in the middle,high dose and cassia twig tuckahoe pill groups was significantly reduced,which was better than the low dose group.The difference was statistically significant(P < 0.05),but there was no statistical difference among the three groups(P > 0.05).The levels of serum IL-2 in the low,medium and high dose groups and the cassia twig tuckahoe pill group were significantly higher than that in the model group(P < 0.05).The content of serum IL-2 factor was significantly increased in the middle and high dose groups of self-proposed anti-tumor prescription prescription,which was better than that of cassia twig tuckahoe pill group(P < 0.05).Conclusion:1.Self-designed anti-tumor prescription has a good therapeutic effect on OEC model rats;2.Self-designed anti-tumor prescription can improve the immunity of the body by reducing the serum levels of TNF-?,VEGF and increasing the levels of IL-2 in the endometriosis cyst model rats.3.Self-designed anti-tumor prescription can reduce the expression of PI3 K inheterotopic lesions of model rats,thus inhibiting the activation of PI3 K signaling pathway and exerting its therapeutic effect on OEC model rats.
