Objective To explore the effect of Gancao Xiexin Decoction on the expression of TNF-?,MCP-1 in serum and the histopathology of intestinal mucosa in antibiotic induced intestinal flora imbalance mice,and further explore the regulatory mechanism of Gancao Xiexin Decoction on intestinal flora imbalance.Methods 48 clean Kunming mice were randomly divided into model group(40mice)And blank group(8 mice).The model group was given Cefradine and gent amicin sulfate mixture by gavage.After the successful establishment of the model,40 mice in the model group were randomly divided into high,medium,low dose group,microecological preparation group and model control group,with 8mice in each group.The high,middle and low dose groups were given 21.84g/k g,10.92g/kg and 5.46g/kg concentrated Gancao Xiexin Decoction respectively,the microecological preparation group was given 0.68g/kg Siliankang,the model control group was given the same dose of normal saline,the blank group was given the same way of gavage,the total administration was 7 days.After 7 days,blood was collected by eyeball extraction,TNF-? and MCP-1 levels in serum were detected by ELISA.After blood collection,the jejunum tissue was taken by laparotomy for about 2 cm,and he staining was carried out to observe the histopathological changes of intestinal mucosa in mice.Results 1.Observe the pathological changes of mice's intestinal mucosa under the microscope: compared with the blank control group,the model control group has obvious pathological changes;compared with the model control group,the high dose group,the middle dose group and the Siliankang group have improved the pathological changes of mice's jejunum;compared with the high dosegroup and the Siliankang group,the middle dose group has significant effect and low effect There was no significant change in the dose group compared with the model control group.2.TNF-?,MCP-1 levels: after the treatment,the high,medium dose and Siliankang groups of ganganganxiexin decoction can reduce the levels of TNF-? and MCP-1 in mice serum,which is statistically significant compared with the model control group(P There was no significant difference between the high dose group and the Siliankang group(P > 0.05);there was no significant difference between the low dose group and the model control group(P > 0.05).Conclusions The high,middle dose and Siliankang groups of ganganxiexin decoction can reduce the pathological changes of intestinal mucosa,promote the recovery of intestinal mucosa,and inhibit the expression of TNF-? and MCP-1 in serum of mice with intestinal flora disorder.The middle dose group of ganganxiexin decoction has a significant effect. |
