Expression Of MMP-19 And Its Correlation With Adhesin In Gastric Adenocarcinoma

Objectives Investigate the expression of matrix metalloproteinase 19(MMP-19)in gastric adenocarcinoma,and analyze its clinical and prognostic significance.Then focus on its correlation with E-cadherin(E-cad)and N-cadherin(N-cad).Methods 64 patients with gastric adenocarcinoma of operation were collected as the study object in the Affiliated Hospital of North China University of Science and Technology from January 2015 to September 2016.Tumor tissue was taken as the observation group,normal gastric mucosa tissue(>5cm from tumor edge)was taken as the control group.The expression of MMP-19,(Human epidermal growth factor receptor-2)HER-2,E-cad and N-cad were detected by immunohistochemistry envision method in two groups.Semi quantitative expression of MMP-19 was detected by Western Blot method in two groups.HER-2 gene(1+ and 2+ in IHC method)was detected by fluorescence in situ hybridization(FISH)method in gastric adenocarcinoma.Results The positive rate of MMP-19 was 57.8% in gastric adenocarcinoma,and 23.4% in normal gastric mucosa(P=0.001)by immunohistochemistry method.The expression of MMP-19 had statistical significant in infiltration depth(P=0.001),tumor thrombus(P=0.004),lymph node metastasis(P=0.001),but there was no statistical significance in different gender,age,differentiation degree,HER-2 expression,cancer nodule expression(P>0.05).Expression of MMP-19 was related to survival time by survival analysis(P=0.020).Negative correlation was found between MMP-19 and E-cad(r=-0.59,P=0.021),but positive correlation was found between MMP-19 and N-cad(r=0.62,P=0.013)by Pearson correlation analyse.Semi quantitative expression of MMP-19 was significantly higher in the observation group than that in the control group by Western Blot method(P=0.016).Conclusions 1 MMP-19 is highly expressed in gastric adenocarcinoma and it involved in tumor formation.2 MMP-19 was statistically significant in the groups with different tumor invasion depth,tumor embolism and lymph node metastasis,and was involved in tumor progression.3 In gastric adenocarcinoma,MMP-19 was negatively correlated with e-cad and positively correlated with n-cad,so as to regulate the adhesion of tumor cells.4 Detection of the expression of MMP-19 in gastric adenocarcinoma may be of certain value in predicting the prognosis.Figure17;Table3;Reference 108...
