Abstract/Summary:
Objective: To investigate the expression of matrix metalloproteinase 9 in pancreatic ductal adenocarcinoma and its relationship with epithelial mesenchymal transformation.Methods: The clinicopathological characteristics of 45 patients with pancreatic ductal adenocarcinoma who underwent radical surgery in our hospital from July,2012 to July,2017 were retrospectively analyzed.The peroxidase method(SP)was used to detect the expression of MMP-9 and E-cad in the tissue samples.Chi-square test and Fisher’s exact test were used to study the correlation between its expression and clinicopathological characteristics.Spearman was used to analyze the correlation between MMP-9 and E-cadherin expression in PDAC.The correlation between the expression of MMP-9 and EMT was further understood.Kaplan-meier method survival curve and COX multivariate analysis were used to investigate the effect of the expression of MMP-9 on the prognosis of patients with PDAC(Bilateral p<0.05 was considered statistically significant).Results:The positive expression of MMP-9 in 45 cases of PDAC was 32(71.1%).The expression of MMP-9 was related to TNM stage and lymph node metastasis,The difference was statistically significant(p<0.05).But the expression of MMP-9 was not related to gender,age,tumor site,,preoperative CA-199 level,tumor size,T stage,tumor differentiation degree,nerve metastasis and vascular invasion.The difference was not statistically significant(p>0.05).The expression of MMP-9 in PDAC was negatively correlated with the expression of E-cad(r=-0.349,p<0.05).Kaplan-meier survival curve showed that the median survival time of patients with high MMP-9 expression was lower than that of patients with low MMP-9 expression,and the difference between the two survival curves was statistically significant.The median recurrence-free survival time of patients with high MMP-9 expression was lower than that of patients with low MMP-9 expression,and the difference between the two survival curves was statistically significant.Multivariate analysis showed that high expression of MMP-9 was an independent risk factor affecting the prognosis of PDAC patients.Conclusion:MMP-9 is overexpressed in PDAC,and its expression may be related to the malignant biological behavior of PDAC.And the high expression of MMP-9 is related to the poor prognosis of patients.The expression of MMP-9 and E-cad in PDAC was negative related,suggesting that MMP-9 may be involved in EMT of PDAC by down-regulating E-cadherin.
