| Objectives To study the specific expression and expression intensity of cyclooxygenase-2(COX-2),programmed death molecular ligand-1(PD-L1)and annexin A2(ANXA2)in benign and malignant parotid tumors.Combined with its gradient expression changes in malignant tumors of different clinical pathological grades and its correlation degree,it can assist in judging the malignant degree of tumors and predicting the prognosis and development of tumors,and provide meaningful pathological reference for differential diagnosis of benign and malignant parotid tumors and clinical prediction and treatment of malignant parotid tumors.Methods Fifty paraffin-embedded specimens of parotid malignant tumors admitted to the Department of Oral and Maxillofacial Surgery of the Affiliated Hospital of North China University of Technology from September 1,2014 to September 2,2019 and treated surgically and confirmed by the Department of Pathology of our hospital were collected as malignant specimen experimental group.Another 50 cases of benign tumor tissue and 50 cases of normal glandular lobe were randomly selected from the same period of tissue specimen archive,respectively as benign specimen experimental group and normal specimen control group.All the patients from the above pathological sections were treated by primary surgery,and didn’t received any radiotherapy,surgical,chemotherapy and hormone treatment before surgery.There were no other tumor lesions in the whole body.Complete medical records and pathological information of the above patients were collected in the medical records department and pathology department of our hospital.Among them,parotid malignant tumors were classified pathologically according to the criteria of Head and Neck Cancer revised by national comprehensive cancer network(NCCN)in June 2019.The representation and intensity of COX-2,PD-L1 and ANXA2 in parotid benign and malignant specimens of experimental group and control group were detected by immunohistochemical SP method.SPSS 21.0 was used to analyze the experimental data.Results 1 The positive representation rates of COX-2 in normal parotid gland,benign parotid gland tumor and malignant parotid gland tumor were 10.0%,42.0% and 90.0% individually.The positive proportion of PD-L1 were 8.0%,46.0% and 82% respectively.The positive representation rates of ANXA2 were 38.0%,58.0% and 78.0%,respectively.Analysis showed,visible,that the positive representation rates of COX-2,PD-L1 and ANXA2 in normal parotid gland,benign parotid gland and malignant parotid gland tissues showed an increasing trend,which got Statistical confirmation.2(1)In parotid gland cancer tissues,the positive expression rates of COX-2 in the four groups of different t classifications are 33.3%,92.9%,100.0% and 100.0%,individually.there are significant differences among the four groups,and the differences are statistically remarkble;The expression rates in patients with long-term smoking and non-long-term smoking were 100.0% and 77.3%,respectively.the difference between the smoker and non-somoker was statistically self-evident.Gender,lymph node and tissue metastasis of tumor were irrelevant to the expression.(2)In parotid gland malignancies tissues,the positive degree of expression of PD-L1 in t-classification tissues were 33.3%,78.6%,90.5% and 100.0%,individually.Obviousness has been proven by statistics among the four groups of different t-classifications.There was no significant difference between the expression rate and lymph node metastasis,distal metastasis,sex and smoking history.(3)In the malignant experimental group,the positive rates of ANXA2 were 62.5%,94.1% and 88.9% respectively in n-classification tissues,and the distinctions,statistically,among the three groups with different n-classifications.92.8% and 59.1% of long-term smokers and nonlong-term smokers respectively,and the difference between the two groups with different individual smoking histories is statistically significant.The positive expression difference of ANXA2 had no statistical significance with gender,tumor size and distal metastasis.3 In the group of malignant experimental,the over-representation index of COX-2 and PDL1 is positively correlated,r= 0.468,obviously;The over-representation rate of PD-L1 and ANXA2 was positively correlated,r=0.364,it has significance.The over-representation index of COX-2 and ANXA2 was positively correlated,r=0.505,it has significance statistics.Conclusions 1 Positive indexes of COX-2,PD-L1 and ANXA2 in healthy parotid pathological section,benign tumors and malignant tumors showed an increasing trend.2.The positive indexes of COX-2 is related to the tumor diameter,and the expression rate is higher in tumor tissues of patients with long-term smoking.The positive index of PD-L1 is related to the size of parotid gland malignant tumor,and the stronger the expression is in tumor tissues with larger diameter.The positive expression rate of ANXA2 is related to lymph node metastasis and smoking history of parotid gland malignant tumors.3.COX-2,PD-L1 and ANXA2 are positively correlated
