In recent years,with the rapid development of network pharmacology technology,it not only provides a new perspective for the modernization of traditional Chinese medicine(TCM),but also provides a new method for the research on the material basis of TCM.Buxue Yimu pill has the functions of promoting blood circulation and regulating meridians,invigorating both qi and blood,removing blood stasis and generating new energy.It is a kind of TCM compound used in clinical treatment of postpartum blood deficiency,blood stasis and other gynecological diseases.However,Its material basis and mechanism have not been reported.We plan to take Buxue Yimu pill and core herb pair“Angelica sinensis-Leonurus japonicus houtt”as the research object,and use the network pharmacology and experimental verification to study the material basis and mechanism of effect of “Angelica sinensis-Leonurus japonicus houtt” in the treatment of type II diabetes mellitus and gynecological diseases,and further study the material effect and synergistic mechanism of Buxue Yimu pill in the treatment of hemorrhagic anemia.The main contents are as follows:(1)Based on the network pharmacology,the effective components and molecular mechanism of “Angelica sinensis-Leonurus japonicus houtt” in the treatment of type II diabetes were revealed.32 anti type II diabetes active components were screened out,and 43 potential targets were enriched.“Angelica sinensisLeonurus japonicus houtt” can to improve the symptoms of complications of type II diabetes by regulating the target to exert hypoglycemic effect.(2)The synergetic regulation mechanism of “Angelica sinensis-Leonurus japonicus houtt” in the treatment of gynecological diseases.72 key active components were screened for the treatment of dysmenorrhea,endometriosis,endometritis,pelvic inflammatory disease,anemia and other diseases,which were enriched to 49 core targets and 15 pathways.Gene GO enrichment and KEGG pathway analysis showed that calcium ?VEGF?MAPK and PI3K-AKT signal pathway were closely related to the pharmacological effects of the drug on tonifying blood,promoting blood circulation and removing stasis,analgesia and anti-inflammatory.The results of ELISA showed that “Angelica sinensisLeonurus japonicus houtt” could down regulate the activities of IL6,VEGF,TNF-? and PTGS2,so as to relieve the symptoms of dysmenorrhea mice.(3)Study on the chemical component of Buxue Yimu pill.62 compounds were identified by GC-MS and UPLC-Q-TOF/MS,including 13 flavonoids,11 terpenes,6 phenylpropanoids,5 aliphatic hydrocarbons,2 alkaloids and 25 others.(4)The synergistic mechanism of Buxue Yimu pill on hemorrhagic anemia.First of all,31 active components were screened by using network pharmacology combined with drug-target structure-activity relationship,which were enriched to52 core targets and 5 core pathways.network analysis found that the pharmacological effect of Buxue Yimu pill in the treatment of hemorrhagic anemia is to promote hematopoiesis and blood circulation,enhance immunity,and play the effect of double tonifying qi and blood.Secondly,the influence of Buxue Yimu pill on the model of hemorrhagic anemia was studied by taking the peripheral blood indexes of the rats with hemorrhagic anemia as the factors.The results showed that Buxue Yimu pill could significantly improve the RBC,HGB and HCT contents.Furthermore,ELISA was used to evaluate the changes of key targets in the blood of rats with hemorrhagic anemia.The results showed that Buxue Yimu pill could significantly reduce EPO,IL6,CSF3,NOS2,VEGFA,PDGFRB and TGFB1.To sum up,“Angelica sinensis-Leonurus japonicus houtt” in the treatment of type II diabetes and gynecological diseases,Buxue Yimu pill in the treatment of hemorrhagic anemia,all of them have the characteristics of multi-component combination,multi-pathway and multi-target coordinated regulation of TCM.
