Objective: To establish the fingerprint spectrum of Rumex japonicus Houtt(RJH)in order to ensure the quality consistency;To further clarify the chemical constituents for its relative effects;To establish a network pharmacology targeting pathway for the treatment of psoriasis;To verify the antifungal activity of the RJH;To indicate the protective effect of the RJH on the UV-B damage of psoriasis skin.Methods: HPLC was used to establish the fingerprint spectrum of five-year-old RJH from different regions;Chromatography and nuclear magnetic resonance(NMR)were used in the chemical research of RJH;The network database and Cytoscape software were employed to study the network pharmacology;The inhibition ring experiment was applied to explore inhibition activity against psoriasis fungi.Protective effect of RJH on psoriatic skin injury against UV-B by enzyme labeled spectrophotometry.Results: The fingerprints spectrum of five-year-old RJH from different areas were established,and 18 common peaks and 6 characteristic peaks were identified.The chemical structures of five constituents from the RJH were identified and assigned systematically for the first time,such as chrysophanol-8-O-β-D-glucopyranoside,aesculin,aloin,aesculin and emodin-8-O-β-D-glucopyranoside.The antifungal activities of different chemical fractions and five compounds from the RJH were investigated,the results showed that ethyl acetate fraction and emodin-8-O-β-D-glucopyranoside had significant anti-fungal activities,and chloroform fraction and Aloin expressed obvious anti UV-B activity,respectively.Conclusion: The establishment of comprehensive and systematic fingerprint spectrum of five-year-old RJH ensured the consistency of application and quality evaluation of RJH;The structural assignments of the chemical constituents were improved,and laid an experimental basis for the furher chemical study of anthraquinones and flavonoids in the root;Network pharmacology research provided target-pathways for anti-psoriasis pharmacology and related mechanism research;The target chemical fractions and monomers were determined by antifungal activity assay,which provided the basis for the further development of new drugs of RJH;The protective effects of chemical components and monomers in RJH laid an experimental foundation for the rational design of safe and effective new drugs against psoriatic skin UV-B damage. |
