This dissertation investigated chemical constituents of the ethanolic extract of Leonurus japonicus, and combined their bioactivities on anti-platelet aggregation of pharmacological models.By using a variety of chromatographic techniques and spectroscopic methods,58compounds (1*-58) were isolated and structurally determined from the parts of D, G, H of the extract. Names and structures of compounds are listed in Table1and Figure1, respectively. Among them, compounds1*-12*are new natural products, compounds1*-6*are novel bis-spirolabdane diterpenoids; compound7*is the dinorlabdane diterpenoid found in genus Leonurus for the first time; compounds13-16are a rare norlupine-type of triterpenoids; and isolates38,39are biphenyl cyclooctene lignans which are first discovered from Labiatae.As for the complexity of diterpenoids in configuration, it is difficult to determine the absolute configuration of the bis-spirolabdane diterpenoids. The absolute configurations of diterpenoids were determined by Mosher’s method, NMR analysis or single-crystal X-ray crystallographic analysis. The study combined with chemical transform method which suggested the configuration of secondary alcohol of the bis-spirolabdane diterpenoids could be determined by Mosher’s method.According to Motherwort’s function of eradicating blood stasis, compounds were isolated for antiplatelet to find some components of a certain activity. The pharmacological test results of bis-spirolabdane diterpenoids showed that C-13of the compounds having R configuration were superior to the S configuration. Suggesting that the activity is closely related to the configuration of C-13.Secondly, the pharmacological of antibacterial activity indicated that the volatile oil of aerial parts was superior to that of seeds. In order to find antimicrobial active ingredients, the essential oils from air dried "Yimucao" and "Chongweizi" were subsequently analyzed by GC/MS systems to identify the differences. Combined pharmacological test with isolation from the volatile oils, our study got a compound with favorable antibacterial activity in the end.
