| OBJECTIVE Reperfusion of acute myocardial infarction (AMI) is the most effective method at present. It has already confirmed that earlier reperfusion of ischemia myocardium can decrease the infarct size, on the other hand the cadiocyte injury may be aggravated,such phenomenon is called ischemia reperfusion injury (IRI). Whether late reperfusion of AMI is benefit to its prognosis? Especially late reperfusion of AMI whether or not exist IRI, has not be reported at present. This research through the late reperfusion of acute myocardial infarction in canine,to detect the influence of late reperfusion of AMI on apoptotic index ,expression of Fas/FasL, and the relationship with oxidative stress reaction.METHODS Thirty healthy canine randomly divided into five groups: control group(C), permanent ischemia group(PI), late reperfusion group(LR), late reperfusion add metoprolol group(LR+M), late reperfusion add carvedilol group(LR+C).All canines silked from the left anterior descending coronary artery.C group didn't ligate the coronary atery;PI group ligated the coronary artery for 6h;LR group ligated the coronary artery for 6h and reperfusion for 6h;LR+M group drenched metoprolol (lmg/kg/d) for seven days,and then ligated the coronary artery for 6h and reperfusion for 6h; LR+C group drenched carvedilol (lmg/kg/d) for seven days,and then ligated the coronary for 6h and reperfusion for 6h.Executed all canines and taken out the heart, Apoptotic index(AI) detected by TdT-mediated dUTP nick end labeling(TUNEL); Fas/FasL gene expression detected by Strept Avidin Biotin Complex (SABC) method;SOD activity detected by xanthine oxidation method,GR activity detected by colorimetry,MDA content detected by barbituric acid... |
