Objective To establish a primary rat hippocampal neuron system in vitro and to further study the mechanism of Sema3F-related hippocampal neuronal growth cone collapse through the expression of HIF-1 ? protein after Sema3 F intervention,so as to provide experimental basis for exploring the mechanism of epilepsy.Method 1.The newborn Wistar rats within 24 hours were selected,and the bilateral hippocampal tissues were selected for cell culture of primary rat hippocampal neurons.2.The primary rat hippocampal neurons cultured for 3 days were extracted and divided into experimental group and control group.In the experimental group,Sema3 F,was added to the cell culture medium to take samples for 0 min,5 min,15 min and 30 min.In the control group,the same concentration of fetal bovine serum was added to the cell culture medium,and the samples were taken from the above four different time points.3.The expression of HIF-1 ?protein in primary hippocampal neurons of experimental group and control group was detected by Western Blot technique.4.The data were analyzed by using SPSS22.0 statistical software.P<0.05 was considered as statistically significant.Results There was no significant change in the expression of HIF-1 ? protein in primary hippocampal neurons in the experimental group after the intervention of Sema3 F,while there was no significant change in the expression of HIF-1 ? protein in primary hippocampal neurons in the control group.Conclusion Sema3 F has no significant effect on the expression of HIF-1 ? protein in primary rat hippocampal neurons.
