Lepista sordida belongs to Fungi,Basidiomycota,Hymenomycetes,Agaricales,Tricholomataceae and Lepista.It has quite a few compositions and multiple pharmacological activities,such as plant growth regulating effect,immunomodulatory effect,hepatoprotective effect,anti-pathogenic bacteria and anti-viral activity,anti-tumor and anti-oxidation activity.In recent years,domestic and foreign scholars have gradually increased their researchs on Lepista sordida,but most of the researchs focus on pharmacological activity,artificial cultivation and resource distribution.There are few studies on chemical compositions.The material basis of the activity can be learnd on the small molecule level by systematic study of chemical composition for Lepista sordida,after mastering the molecular structure of the compound in the fruiting body of Lepista sordida,specific pharmacological activities of different molecular structures and structure activity relationship can be explored,which is conducive to accelerate the industrialization process of Lepista sordida.1.Study on the chemical compositios of Lepista sordida.Six monomer compounds were isolated and identified by silica gel column,medium pressure column chromatogr aphy,gel column chromatography and HPLC,including five sterol compounds,which w ere 3β,5α,9α,14α-tetrahydroxy-(22E,24R)-ergosta-7,22-dien-6-one(1),3β,5α,9α-trihydroxy-(22E,24R)-ergosta-7,22-dien-6-one(2),5α,6α-epoxy-(22E,24R)-ergosta-8(14),22-diene-3β,7α-diol(3),cerevisterol(4),ergosterol(5),the last one was dibutyl phthalate(6).Compound 1,2,4,and 6 were isolated from Lepista for the first time.2.Extraction process of sterol components in Lepista sordida was optimized by re sponse surface method.It was determined that optimal solution to material ratio was 10:1,optimal extractive temperature was 85℃,optimal extractive time was 3.5 h,and optimal ethanol concentration was 95% through single factor experiment.On this basis,sterols extraction process was optimized by response surface method,the result showe d that the optimal solution to material ratio was 10.32:1,optimal extractive temperature was 84.33℃,optimal extractive time was 3.66 h,and predicted extraction rate of ste rols was 0.136%.Verified value was 0.145% using the predicted extraction conditions,which was not much different from the predicted digital.
