Objective:To study the effect of propofol on cognitive function in young obese rats and to analyze its relationship to the expression of?-amyloid protein,Tau protein and p-Tau protein in hippocampal tissue.Methods:One hundred and forty 3-week-old male Sprague-Dawley?SD?rats were randomly divided into two groups:normal diet group?n=40?and high-fat diet group?n=100?.After being fed with basic feed for four weeks,the normal group was randomly divided into two groups:normal lipid emulsion solvent group?NL?and normal propofol group?NP?,with 20rats in each group.After the high-fat diet group was fed with the high-fat diet for four weeks,the body weight of the rats was equal to the average body weight of the basic diet-fed group plus 1.4 times the standard deviation,which was regarded as a successful modeling of the obesity model.The obesity rats were then randomly divided into two groups:obesity lipid emulsion solvent group?OL?and obesity propofol group?OP?,with 20 rats in each group.The normal lipid emulsion solvent group and obesity lipid emulsion solvent group were given intraperitoneal injection of the same amount of 10m L·kg-1 lipid emulsion,and the normalpropofol group and obesity propofol group were given intraperitoneal injection of the 100mg·kg-1 propofol.All rats were injected continuously for 7 days,once a day.Cognitive function was detected by water maze test;the neuronal apoptosis in hippocampus were detected by TUNEL method;the expression of?-amyloid protein?A??,Tau protein and p-Tau proteinin hippocampal tissue were detected by Western blot.Results:There was no significant difference in the initial body weight between the normal diet group and the high-fat diet group.After four weeks of feeding,rats in obesity group gained more body weight than the normal diet group?P<0.01?.According to the standard of obese there are 40 the models rats in obesity,the obesity rate of the rats is 40%.?2?Results of the water maze experiment showed:Compared with the NP group and OL group,the escape latency in OP group was significantly longer on the 3nd to 5th day?P<0.05?.Compared with the NL group,the escape latency in the NP group and OL group was significantly longer on the 1nd to 2nd day?P<0.05?.Compared with the NP group and OL group,the times of crossing the platform in the OP group was less?P<0.05?and the target quadrant time?P<0.05?in the OP group was shorter?P<0.05?.?3?Results of the TUNEL staining showed:Compared with the NP group and OL group,the neuronal apoptosis in the OP group was significantly increased?P<0.01?.?4?Results of Western blot analysis showed:Compared with the NP group and OL group,the expression of A?protein in the OP group was significantly increased?P<0.05?.Compared with the NP group and OL group,theexpression of p-Tau protein in the OP group was significantly increased?P<0.05?.Compared with the NP group and OL group,the expression of Tau protein in the OP group was significantly increased?P<0.05?.There were no significant differences in the A?,Tau and p-Tau protein between NL group,NP group and OL group.Conclusion:Propofol anesthesia can reduce the cognitive function in young obese rats,it may be related to increase the expression of?-amyloid protein and Tau protein.
