Objective:To study the effect of acupuncture at Back-Shu and Front-Mu Points on the treatment of functional constipation and its effect on gut microbiota.Explore the mechanism of acupuncture and provide new evidence for the theory of Back-Shu and Front-Mu Points.Methods:Thirty Sprague-Dawley male rats were allocated to blank group(Group BK),model group(Group BM),Back-Shu point group(Group B.S),Front-Mu point group(Group B.M)and Shu-Mu points group(Group B.SM)using a random number table,for each group with six rats.Functional constipation model was established by gastric infusion with compound diphenoxylate for 14 days.When the model was finished,electroacupuncture Group B.S(ST 25),Group B.M(BL 25)and Group B.SM(ST 25 + BL 25)for 20 minutes/session,one session a day,a total of 10 day.Rats in Group BK and Group BM were bundled in the same way as electroacupuncture group but with no needling.After the intervention,stool feces were collected in aseptic condition.Intestinal flora was detected by 16 S r DNA sequencing technology.Results:1.Compared with Group BK,the number of defecations in Group BM was reduced,and reached statistical significance(P <0.05).The number of defecations in Group B.S,Group B.M,and Group B.SM were increased when compared with Group BM.however,only Group B.SM achieved statistical significance(P <0.01).No statistical significance was achieved among Group B.S,Group B.M,and Group B.SM.Compared with Group BK,the fecal water content in Group BM was lower,and the difference reached statistical significance(P <0.05).Compared with Group BM,the fecal water content in Group B.S,Group B.M,and Group B.SM were increased with Group B.M and Group B.SM achieved statistical significance(P <0.05).There was no statistical significance among Group B.S,Group B.M,and Group B.SM.2.There was no significant difference in Alpha diversity between Group BK and Group BM.Beta diversity analysis showed that the microbial community in Group BM was significantly different when compared with Group BK in the PC1 dimension.The relative abundances of Euryarchaeota,Verrucomicrobia,Deltaproteobacteria(Desulfovibrionales,Desulfovibrionaceae),S24-7 and Veillonella were increased with statistically significant differences(P <0.05).The relative abundances of Bacilli(Lactobacillales,Lactobacillaceae,Lactobacillus)were decreased with statistical significance(P <0.05)3.There was no significant difference in Alpha diversity among Group BM,Group B.S,Group B.M and Group B.SM.Beta diversity analysis showed that the microbial community in Group B.SM was significantly different when compared with Group BM.Compared Group B.S to Group BM,the relative abundance of the Paraprevotellaceae(CF231)was decreased,and the difference achieved statistical significance(P <0.05).The relative abundance of Peptococcaceae(rc4-4),Prevotellaceae,Helicobacteraceae(Helicobacter),Phascolarctobacterium was increased with statistical significance(P <0.05);Compared Group B.M to Group BM,the relative abundance of Gammaproteobacteria,Paraprevotellaceae,S24-7 was decreased,and the difference reached statistical significance(P <0.05).The relative abundance of Bacilli(Lactobacillales,Lactobacillaceae,Lactobacillus)and rc4-4 was increased,the difference achieved statistical significance(P <0.05);Compared Group B.SM to Group BM,the relative abundance of Gammaproteobacteria was decreased and reached statistical significance(P<0.05).The relative abundance of Prevotellaceae and Paraprevotellaceae(rc4-4)was increased,and the difference was statistically significant(P <0.05).Conclusions:1.Rats with functional constipation existed weakened gut motility,and disordered intestinal flora.2.Electroacupuncture at Front-Mu Point(BL 25)and Back-Shu Point plus FrontMu Point(ST 25 + BL 25)can regulate the gut motility,increase the number of defecation and fecal water content in rats with functional constipation.Electroacupuncture at Back-Shu point(ST 25),Front-Mu Point(BL 25)and Back-Shu Point plus Front-Mu Point(ST 25 + BL 25)can adjust intestinal flora of functional constipation Rat.3.The enhancement of gut motility by electroacupuncture at Front-Mu Point(BL 25)may be related to increasing the abundance of Lactobacillus and reducing the abundance of ?-proteobacteria.The enhancement of gut motility by electroacupuncture at Back-Shu Point plus Front-Mu Point(ST 25 + BL 25)may be related to increasing the abundance of Prevotellaceae,and reducing the abundance of Gammaproteobacteria. |
