Objective: To observe the effect of Baishen zinc oxide paste on anti-inflammatory and expression of AQP3 and FLG in guinea pigs with subacute eczema,and to explore the anti-inflammatory effect of Baishen zinc oxide paste on subacute eczema and its regulation on skin barrier function,so as to provide basis for further elucidating the molecular biology of Baishen zinc oxide paste on subacute eczema.Methods: Thirty two guinea pigs were randomly divided into blank group,model group,Baishen Zinc oxide paste group and mometasone furoate group,8 in each group.One day before the formal experiment,except for the blank group,the other three groups of guinea pigs were depilated on the back,with an area of about 2cm × 2cm.On the day of experiment,except for the blank group,all the remaining groups were sensitized with 5% DNCB acetone solution 25 ul on the back of guinea pigs.After 2 weeks,each guinea pig was exposed to 100 UL of 0.1% DNCB acetone solution on the inner side of the right ear,and the stimulation process was repeated once a week for 4 consecutive times to establish the subacute eczema model.72 hours after the fourth excitation,except for the blank group,each group was given corresponding drugs twice a day for 14 days.At the end of the treatment,the guinea pigs were killed by dislocating the cervical vertebrae,and the drugs in the right ear were wiped off with cotton swabs.Six guinea pigs were taken from each group and their ears were cut off immediately along the baseline of the two ears of the guinea pigs The right ear tissue of the guinea pig group was obtained by the perforator.When the material was taken,it should be kept the same position of the ear piece,and the swelling degree of the right ear of the guinea pig should be measured accurately.Then,the right ear tissue of the guinea pig in each group was placed in the prepared fixed solution of the tissue sample for fixation,and the pathological tissue section was made,and he staining was used to calculate the number of inflammatory cells infiltrated into the real skin,The expression levels of AQP3 and FLG were detected by immunohistochemistry,and the data were analyzed statistically.Result: 1.After the model was established successfully,the right ear of guinea pig showed subacute eczema,with different degrees of diffuse redness,small papule,slight erosion,blood scab,scratches,some of the surface rough and dry,covered with scales.After 14 days of administration,the lesions in the model group showed no significant improvement.The redness and swelling of Baishen Zinc oxide paste group and alosone group were significantly reduced,the scab of blood was off,and pigmentation was seen in the slightly rough part of the skin,which was close to the normal skin by naked eyes.2.Compared with the blank group,the epidermis of guinea pig's right ear in the model group was obviously thickened,with incomplete keratosis,a small amount or more of acanthocytes or basal cells degenerated or necrotic under the cuticle,and a large number of neutrophils or lymphocytes infiltrated in the stroma;there was hemorrhage and red blood cells oozed out;compared with the model group,the epidermis of guinea pig's right ear in the alosone group was thickened in varying degrees,and the stroma was interstitial A small number of inflammatory cells were scattered occasionally;compared with the model group,the epidermis of the right ear of the guinea pig in the Baishen Zinc oxide paste group was thinner,no obvious thickening was found,and a small number of inflammatory cells were infiltrated in the interstitium.3.Compared with the blank group,the swelling degree of the right ear in the model group increased significantly(P < 0.05);compared with the model group,the swelling degree of the right ear in the Baishen Zinc oxide paste group and the alosone group decreased significantly(P < 0.05).4.Compared with the blank group,the density of inflammatory cells in the model group increased significantly(P < 0.05);Compared with the model group,the density of inflammatory cells in the ear tissue of the rats in Baishen Zinc oxide paste group and alosone group decreased significantly(P < 0.05).5.Compared with the blank group,the AQP3 protein content in the right ear of guinea pigs in the model group increased significantly(P < 0.05);compared with the model group,the AQP3 protein content in the right ear of guinea pigs in the Baishen zinc oxide paste group and the alosone group decreased significantly(P < 0.05).Compared with the blank group,the FLG protein content in the right ear of guinea pigs in the model group was significantly lower(P < 0.05);compared with the model group,the FLG protein content in the right ear of guinea pigs in the Baishen zinc oxide paste group and the alosone group was significantly higher(P < 0.05).Conclusion: 1.Baishen Zinc oxide paste can make subacute eczema basically disappear and play a therapeutic role.2.After being treated with Baishen Zinc oxide paste,the thickness of inner epidermis and inflammatory reaction of right ear were obviously reduced,which suggested that Baishen Zinc oxide paste had anti-inflammatory effect.3.The swelling was significantly reduced after treatment witht Baishen Zinc oxide paste.It is suggested that Baishen Zinc oxide paste has the effect of detumescence.4.After the treatment of Baishen Zinc oxide paste,the content of AQP3 protein decreased and the content of FLG protein increased significantly.The results showed that the function of repairing skin barrier could be achieved by down regulating the expression of AQP3 protein and up regulating the expression of FLG protein.
