Objectives To observe the dynamic changes of T lymphocyte,natural killer cells,soluble interleukin-2 receptor(s IL-2R),interleukin-6(IL-6)and immunosuppressive acidic protein(IAP)in peripheral blood of patients with colorectal cancer by transfusion of allogeneic leukocyte-free blood,so as to explore the effect of transfusion on the immune function of patients with colorectal cancer during perioperative period.Methods 64 patients with rectal cancer or colon cancer ASAI ? II admitted to our hospital from May 2016 to May 2017 were enrolled in the study.They were divided into allogeneic leukocyte-free blood transfusion group(group I,300?450 ml of leukocyte-free blood transfusion)and non-transfusion group(group II),32 cases in each group.Peripheral venous blood was collected before anesthesia induction,before blood transfusion,1,3 and7 days after operation.IAP was measured by one-way immunodiffusion method,s IL-2R and IL-6 were measured by double-antibody sandwich ELISA,and CD3+,CD4+,CD8+,CD4+/CD8+ ratio and CD56+ were measured by flow cytometry.The results were analyzed statistically.Results 1.There was no significant difference between the two groups in general condition and operation condition(P>0.05).2.The number of T cell subsets and NK cells before transfusion were decreased in the two groups before the induction of anesthesia(P<0.05).On the 1st day after transfusion,the number of CD3+ and CD4+ in the two groups decreased compared with that before transfusion(P<0.05).3.After operation,CD3~+,CD4~+,CD4~+/CD~8+and CD56~+were all significantly recovered,but there was no statistically significant difference between the two groups(P<0.05).4.The levels of IAP,IL-6 and s IL-2R in the two groups were increased compared with those before induction of anesthesia(P<0.05).Conclusions Perioperative infusion allogeneic leukocyte-free blood cell suspension had a mild inhibitory effect on the immune function of patients with colorectal cancer,and the patients recovered quickly after operation.Therefore,in the current period of blood shortage,we should try our best to avoid transfusion and use the limited blood resources on the patients who really need it,so as to make the clinical use of blood truly scientific and reasonable.Figure11;Table12;Reference 91...
