Objectives This study attempts to investigate the relationship between the levels of comparative gene identification-58(CGI-58)and lipoprotein lipase(LPL)in placenta and preeclampsia,analyzing their clinical relationship.Methods 37 cases of pre-eclampsia pregnant women delivered in the Affiliated Hospital of North China University of Science and Technology from September 2018 to July 2019 were selected as the case group,40 healthy pregnant women without pregnancy complications and complications in the same period as the control group To collect the clinical data of two groups of pregnant women.The levels of CGI-58 and LPL in placenta were measured by immunohistochemistry.The relative expression of CGI-58 and LPL m RNA and protein in placenta were measured by real-time fluorescence quantitative PCR(RT-PCR)and Western blotting respectively.Independent sample t test is used for measurement data.Chi square test was used to compare the measurement data between the two groups.Pearson linear correlation analysis was used to compare the relationship between the two variables,and multivariate logistic regression model was used for multivariate analysis.Results 1 Compared with the blood lipid of healthy pregnant women,TG,LDL-C,apo B,AI were considered to be significantly higher in patients with preeclampsia,HDL-C,Apo A were considered to be significantly lower in patients with preeclampsia,the difference was intense statistically significant(P<0.05).2 Immunochemistry dyeing in placenta tissue showed that the positive expression rate of CGI-58 protein in case group was lower than that of control group,the difference was intense statistically significant(c2=8.016,P<0.05);The positive expression rate of LPL protein in placental tissue of case group was significantly lower than that of control group(c2=10.970,P<0.05).3 The results of RT-PCR showed that the relative expression of CGI-58 m RNA in preeclampsia placenta(0.51±0.13)was lower than that in the control group(1.00±0.00),the difference was statistically significant(t=6.297,P<0.05).The relative expression of LPL m RNA in preeclampsia placenta(0.49±0.10)was lower than that in the control group(1.00±0.00),the difference was statistically significant(t=8.431,P<0.05).4 Western blotting in placenta tissue showed that the relative expression of CGI-58 protein in the case group(0.63±0.19)was lower than that of the control group(0.87±0.12),the difference was statistically significant(t=2.837,P<0.05).The relative expression of LPL protein in placental tissue of case group(0.45±0.04)was lower than that of control group(0.74±0.10),the difference was statistically significant(t=4.815,P<0.05).5 Through linear correlation analysis,we found that there was a positive correlation between CGI-58 protein and LPL protein(r=0.602,P<0.05);CGI-58 protein was positively correlated with serum HDL-C(r=0.625,P<0.01),negatively correlated with TG and apo B(r=-0.840,P<0.01;r=-0.514,P<0.05);LPL protein was positively correlated with HDL-C(r=0.524,P<0.01),negatively correlated with TG and AI index(r=-0.659,P<0.01;r=-0.496,P<0.01).Conclusions 1 The expression of CGI-58 and LPL in placenta of preeclampsia patients was significantly decreased,and the expression of CGI-58 and LPL was positively correlated.2 The expression of CGI-58 and LPL in placenta is related to the pathogenesis of preeclampsia.When the expression level of CGI-58 and LPL is decreased,the risk of preeclampsia is increased.3 CGI-58 protein and LPL protein levels in placenta are related to maternal lipids.Figure 6;Table 15;Reference 139...
