Expression Of Mitochondrial Fusion Protein 2 In Peripheral Blood And Placental Trophoblasts Of Patients With Preeclampsia | | Posted on:2022-12-16 | Degree:Master | Type:Thesis | | Country:China | Candidate:N N Li | Full Text:PDF | | GTID:2504306761457324 | Subject:Oncology | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the expression of mitochondrial fusion protein 2(mitofusin,Mfn2)in peripheral blood and placental trophoblasts of patients with early onset preeclampsia(eo PE)and late onset preeclampsia(lo PE),and to determine that the gene may be used as a target for diagnosis and treatment of preeclampsia.To investigate the difference in the expression of mitochondrial fusion protein 2 between early-onset preeclampsia and late-onset preeclampsia,and to provide experimental data for exploring the pathogenesis between them.Method:1.The placental tissue and serum of 28 patients with early-onset preeclampsia,26 patients with late-onset preeclampsia and 30 pregnant women without obstetrical complications were collected from the second Hospital of Jilin University.The grouping standard was based on the 9th edition of Obstetrics and Gynecology edited by Xie Xin et al.All the parturients in the three groups were singleton pregnancy and natural pregnancy,and there were no complications such as heart disease,diabetes,hematopathy,thyroid dysfunction and so on.2.The expression of Mfn2 in peripheral blood was determined by ELISA method,and the expression of Mfn2 in placental trophoblasts was detected by Westernblot and immunohistochemistry.3.The expression of Mfn2 in peripheral blood determined by ELISA was statistically analyzed by Graph Pad Prism9.0.0.4.The protein bands detected by Western blot were analyzed by Image J software.The expression of Mfn2 was standardized by β-Actin expression and statistically analyzed by Graph Pad Prism 9.0.0.5.The average optical density(AOD)of Mfn2 positive signals in immunohistochemical images was analyzed by Image J software and statistically analyzed by Graph Pad Prism9.0.0.6.The correlation between the expression of Mfn2 in placental trophoblast,peripheral blood and blood pressure was analyzed by software SPSS26.0.Result:1.There was no significant difference in gestational frequency and age among28 patients with early-onset preeclampsia,26 patients with late-onset preeclampsia and 30 pregnant women without obstetrical complications delivered by cesarean section in the second Hospital of Jilin University(P>0.05).There was significant difference in gestational weeks(P<0.0001).The systolic blood pressure and diastolic blood pressure in patients with preeclampsia were significantly higher than those in normal group(P<0.0001),and the systolic blood pressure and diastolic blood pressure in patients with eo PE were significantly higher than those in patients with lo PE(P<0.0001).2.The expression of Mfn2 in peripheral blood of patients with preeclampsia was significantly higher than that of normal controls.The expression of Mfn2 in patients with eo PE was significantly higher than that in patients with lo PE.3.The expression of Mfn2 in placental trophoblasts of patients with eo PE was significantly lower than that of normal group and lo PE group by Western blot and immunohistochemistry.4.At the same time,Western blot and immunohistochemical detection showed that the expression of Mfn2 in placental trophoblasts of patients with lo PE was lower than that of normal controls,but the difference was not statistically significant.5.Pearson correlation analysis showed that the expression of Mfn2 in peripheral blood was positively correlated with blood pressure,while the expression of Mfn2 in placental trophoblasts was negatively correlated with blood pressure.Conclusion:1.The expression of Mfn2 in peripheral blood of preeclampsia is increased,and the increased expression may be related to the occurrence and progression of preeclampsia.It can be used as a clinical index to evaluate the occurrence and severity of preeclampsia.2.Mfn2 expression in the mitochondria of placental trophoblasts of preeclampsia suggests that the pathogenesis of early-onset preeclampsia may be different from that of late-onset preeclampsia,which needs to be further confirmed. | | Keywords/Search Tags: | Early-onset preeclampsia, late-onset preeclampsia, mitochondrial fusion protein 2, Elisa, Western blotting, immunohistochemistry | PDF Full Text Request | Related items |
