Osteoporosis?OP?is a kind of bone disease with bone loss and deterioration of bone structure,which leads to the increase of fracture susceptibility.Around the world,the incidence rate of OP has exceeded25%.With the aging of the population,OP has become one of the diseases with high incidence rate,high mortality rate and high medical expenses.It is a serious public health problem that will seriously harm the health of the elderly.Therefore,it is of great social and economic significance to strengthen the study of this disease.In order to find the active compounds against OP,the chemical constituents of three medicinal plants,Millettia speciosa Champ.,Daphne papyracea wall.,and Lycopodium fargesii Hart.,were studied systematically.Sephadex LH-20,MCI gel CHP-20P,Silica gel column and semi-preparative HPLC were used to systematically separate the extracts of the three plants.The structures of 53 compounds were identified by 1H-NMR,13C-NMR,DEPT and HR-ESI-MS.Combinedwith the osteoclast activity screening model,some compounds were found to have good biological activity.In this study,11 compounds were identified in Millettia speciosa Champ.as millettiaosa A?N-1?,maackiain?N-2?,medicarpin?N-3?,lupeol?N-4?,?-sitosterollinoleate?N-5?,?-sitosterol?N-6?,?-monopalmitin?N-7?,?2S?-1-O-heptatriacontanoyl glycerol?N-8?,vanillin?N-9?,methyl hydrogen succinate?N-10?and 1-octanol?N-11?.Compounds N-5 and N-10 were firstly isolated from the genus Millettia.21 compounds were identified in Daphne papyracea Wall.as?-amyrin?B-1?,oleodaphnone?B-2?,sesamin?B-3?,velutin?B-4?,vanillin?B-5?,daphneolone?B-6?,1,3,5-trimethoxybenzene?B-7?,neodaphnoretin?B-8?,?24S?-stigmasterol-4-en-3-one?B-9?,?-sitosterol?B-10?,28?-homo-?-amyrin acetate?B-11?,?-tocopherolquinone?B-12?,pinoresinol?B-13?,nortrachelogenin?B-14?,syringaresinol?B-15?,isolariciresinol?B-16?,ethyl laurate?B-17?,?-amyrone?B-18?,pentacosane?B-19?,squalene?B-20?,daphnetin?B-21?.Compounds B-8,B-9,B-11 and B-12 were firstly isolated from the genus Daphne.21 compounds were identified in Lycopodium fargesii Hert.as1,20-eicosanediol?M-1?,?-onocerin?M-2?,tricosane?M-3?,serrat-14-en-3?,21?,24-triol?M-4?,26-nor-8-oxo-?-onocerin?M-5?,glycerol monolinoleate?M-6?,glyceryl linolentate?M-7?,serrat-14-en-3?,21?,24-triol?M-8?,ferulaic acid?M-9?,vanillic acid?M-10?,4-hydroxybenzaldehyde?M-11?,3-hydroxy-4-methoxybenzoic acid?M-12?,trans-4-hydroxy-2-nonenoic acid?M-13?,decane-1-thiol?M-14?,4-hydroxy-3-methoxyzimtaldehyde?M-15?,8-Acetoxy-5-hydroxyumbelliprenin?M-16?,3-?4?-formylphenoxy?-4-methox-ybenzaldehyde?M-17?,apteniol G?M-18?,2-hydroxy-1-?4-hydroxy-3-methoxyphenyl?-propan-1-one?M-19?,genkwanin?M-20?and apigenin?M-21?.Compounds M-1,M-6,M-12,M-13,M-14,M-15,M-16,M-17,M-18 and M-19 were firstly isolated from the genus Lycopodium.The activity of the separated compounds was studied.Among of them,compounds N-2,N-3,N-4,N-6,B-2,B-5,B-6,B-12,B-13 and B-15 exhibited potent inhibition.Compounds N-2,B-6 and B-12 were firstly reported inhibitory effects on osteoclastic cell formation activity.
