Millettia Speciosa Champ. is a popular plant with dual purposes of food and herb in the South of china. So far, there are few reports about the chemical compositions and the pharmacological effects of Millettia Speciosa Champ..In our researches, polysaccharide was extracted and purified from Millettia Speciosa Champ..Main physico-chemical properties of the polysaccharide were determined,and the structures of the polysaccharide were characterized. Its pharmacological effects on antioxidation, anti-inflammatory, anti-cancer and so on were studied. These studies may provide basic datas for the exploitation and the applicaton of Millettia Speciosa Champ..They also provide theoretical and experimental evidences for the clinical application of Millettia Speciosa Champ..The major effective components of Millettia Speciosa Champ. are in the hydrosoluble part, which have many water-soluble polysaccharides. Millettia Speciosa Champ. were cooked by water and precipitated by ethyl alcohol. The filtrates were purified and the remaining solutions were freezed and dried. Polysaccharides of Millettia Speciosa Champ. were obtained and the main compositions of polysaccharide were obtained by glucosan gelatin column chromatographic analysis. Antioxidation capacity and radical scavenging activity of the polysaccharides from Millettia Speciosa Champ. were investigated by detecting anti-lipid peroxidation, hydroxy radical scavenging activity and DPPH·radical scavenging activity. The effects of polysaccharide on mouse lymphocyte proliferation stimulated by Con A were investigated by flow cytometry technique. The effects of polysaccharide on the models of acute inflammatory reactions of xylene induced mouse ear edema were studied and the effects of polysaccharide on Hela in vitro growth were observed by microscope and MTT method. Models of immunosuppressed mice infected with cyclophosphamide were performed to study the effects of polysaccharide on hemolysin production leve and immune organs index. Elements in Millettia Speciosa Champ. were determined by ICP atomic emission spectroscopy. Elements'composition in the polysaccharide was determinated by elemental analyzer. The monosaccharide compositions of polysaccharide were identified by ion chromatograph. Thecharacteristic absorption of the main constituents of polysaccharide was detected by infrared spectrum and the molecular weight scope of the main constituents of polysaccharide was determinated by gel chromatography.The contents of 19 kinds of elements in Millettia Speciosa Champ. were determined in the experiment, among which the contents of Ca, K, Mg, Al, Fe and Mn were abundant. The elements composition in the polysaccharide were C 39.45%, H 6.27%, and N 0.49% respectively. The monosaccharide compositions of hamnose, galactose, glucose, mannose and fructose in the polysaccharide were 5.2, 59.9, 4717.9, 25.7 and 1345.0 mg/kg respectively. The main compositions of polysaccharide (MSP1) were isolated by glucosan gelatin column G-75. There were typical characteristic absorption peaks of polysaccharide in MSP1. Its number average molecular weight (M_n) was 6.64×10~3, weight average molecular weight (M_w) was 9.05×10~3, and distribution width (D) was 1.36. Effects of polysaccharide on T lymphocyte proliferation in mouse lymph node stimulated by Con A were related to the concentration of polysaccharide. 160μg/ml, 0.8mg/ml , 4mg/ml and 20mg/ml concentrations of polysaccharide restrained the T lymphocyte proliferation of mouse with significant dose-dependent, while 5, 25, 50 and 125μg/ml concentrations of polysaccharide promoted the T lymphocytes proliferation of mouse without significant dose-dependent. Polysaccharide of Millettia Speciosa Champ. effectively inhibited the growth of Hela cell; significantly suppressed xylene induced mouse ear edema (P<0.01 or P<0.05); markedly promoted serum hemolysin production (P<0.01 or P<0.05) and increased immune organs index. Experiments revealed the order of the abilities of·OH scavenging and anti-lipid peroxidation (from strong to weak) were: water extract > ethanol extract> polysaccharide, while the ability of DPPH·scavenging (from strong to weak) was: ethanol extract> water extract > polysaccharide.The polysaccharide of Millettia Speciosa Champ. have activities on anti- inflammatory and can improve immunefunction. It can effectively scavenge free radical and antioxidate,either. The polysaccharide of Millettia Speciosa Champ. was worthy of being further exploited and utilized, It was a kind of natural product with many kinds of pharmacological activities and good health care function.
