Objective:In this study,by comparing the differences in the expression of sB7-H3 in the serum of breast cancer patients and healthy patients,and at the same time analyzing the correlation between sB7-H3 and postoperative pathological parameters of breast cancer patients and the expression of B7-H3 in cancer tissues,in order to preliminary explore the clinical value of sB7-H3 expression in the serum of breast cancer patients.Method:1.sB7-H3 levels in serum of 106 patients with invasive breast cancer and 43 healthy people obtained from the health examination center were detected by ELISA.Differences in sB7-H3 expression between the two groups were compared,and the relationship between serum sB7-H3 levels in breast cancer patients and pathological data parameters was analyzed.ROC curve was plotted to obtain the optimal cut-off value for breast cancer diagnosis.2.To obtain the samples of breast cancer tissues and paracancerous tissues from the last 30 of 106 patients with invasive breast cancer,the expression levels of B7-H3 in the cancer tissues and paracancerous tissues were detected by immunohistochemistry,and the differences between the expression levels of B7-H3 in the cancer tissues and paracancerous tissues were analyzed and compared.3.To analyze the correlation between sB7-H3 expression level in serum of breast cancer patients and B7-H3 expression level in corresponding cancer tissues.Results:1.The expression of sB7-H3 in the serum of breast cancer patients was 26.27 ±4.21ng/ml,the expression of sB7-H3 in healthy people was 19.44 ± 4.91ng/ml,t=8.53,P<0.05,the difference was obvious.The optimal cut-off value of sB7-H3 in the serum of breast cancer and healthy people is 21.73ng/ml.Univariate analysis of the relationship between the expression of sB7-H3 in serum and pathological parameters of breast cancer patients,we found that the expression of sB7-H3 in breast cancer patients with high pTNM(stage ?)was lower than that of stage(stage ? and ?)sB7-H3 expression in the serum of the patients,t=-2.219,P<0.05,the difference was statistically significant.Compared with the serum of patients without lymph node metastasis,the expression of sB7-H3 in patients with lymph node metastasis was significantly increased,t=-2.205,P<0.05,the difference was statistically significant.The expression of sB7-H3 in patients with TILS(tumor infiltrating lymphocytes)?5%is higher than that in patients with TILS>5%,t=2.067,P<0.05,the difference is statistically significant,while in other In the analysis of pathological parameter grouping,P>0.05,no obvious difference was found.2.Immunohistochemical staining showed that B7-H3 was mainly expressed in the cell membrane and cytoplasm,and the surrounding matrix was also strongly expressed.Non-parametric rank sum test analysis showed that B7-H3 was significantly different in the expression level of cancer tissues and adjacent tissues.,Z=-3.908,P<0.05.3.Spearman rank correlation analysis showed that there was a positive correlation between the expression level of sB7-H3 in the serum of breast cancer patients and the expression level of B7-H3 in the corresponding cancer tissue.The correlation coefficient was 0.526,P<0.05.The parallel line test of logistic regression analysis P=0.124>0.05,regression analysis P<0.05,estimated value=-2.292,OR value=exp(-2.292)=0.101,indicating that the newly diagnosed patients with higher expression of sB7-H3 are expected to express B7-H3 expression level of cancer tissue than patients with low expression higher.Conclusion:1.The expression level of sB7-H3 in the serum of breast cancer patients was significantly higher than that of healthy people,and was related to some pathological parameters,suggesting that sB7-H3 may be a potential tumor marker for auxiliary diagnosis of breast cancer and may be involved in the occurrence and progression of breast cancer.2.B7-H3 is highly expressed in breast cancer cells and surrounding matrix in breast cancer patients,and B7-H3 in cancer tissues expression is significantly higher than that of adjacent tissues,suggesting that we carry out imaging diagnosis and drug therapy targeting B7-H3 may become a new method to improve the diagnosis and treatment of breast cancer.There is a positive correlation between sB7-H3 in patients'serum and B7-H3 expressed in cancer tissues,supporting the view that sB7-H3 in patients' serum may come from tumor cell membranes with positive B7-H3 expression.3.In this study,we found that sB7-H3 in serum can reflect the expression of B7-H3 in cancer tissues.At the same time,based on literature reports that B7-H3 expression in breast cancer tissues is related to prognosis,the above results suggest that in the future,we can further investigate whether the expression level of sB7-H3 in patients' peripheral serum can prompt patients' prognosis.
