Objective: To investigate the effect of propofol on blood hypercoagulability in burned rats.Methods:Thirty male SD rats at the age of 10 weeks were selected,and their body weight was controlled at 220±10 g.All operations and animal treatments were performed according to experimental animal guidelines and standards.The experiment was started 7 days after acclimation,and rats were grasped for 5 minutes every day during the acclimation period to reduce the impact of irrelevant stress stimulation on subsequent experimental operations.Rats were randomly divided into three groups(n = 10): propofol + burn(A)group,simple burn(B)group and blank control(C)group.Rats in groups A and B were anesthetized with 1% sodium pentobarbital 45 mg / kg intraperitoneally.The rat model of third-degree burn was built according to the laboratory.After being stabilized for 1 hour,group A received intraperitoneal injection of 40ml/kg sodium lactate linker solution,and 20mg/kg propofol with A concentration of 1% was injected via femoral vein,followed by continuous pumping at A rate of 10mg/kg/h,and maintained infusion until the end of the experiment.In group B,1 ml / kg / h physiological salt water pump was used to replace the fluid supplementation,and 1% pentobarbital sodium was added separately to ensure that the rats did not show body movement during the experiment.Group C was replaced with a saline injection of 1 ml / kg / h.Changes of endothelin-1(ET-1),platelet aggregation rate(PAg R),and platelet membrane glycoprotein CD62 P were detected at 1h(T1),3h(T2),and 5h(T3)after burns.Results: In the detection of endothelin 1(ET-1)levels in burned rats,it was found that:The level of et-1 in the group of propofol + burn(A)decreased significantly at the time points of T2 and T3,but it was still higher than that in the normal group(P < 0.05).In the detection of the platelet aggregation rate(PAg R)level in post-burn rats,it was found that:The level of PAg R in group A gradually decreased at time points T2 and T3,which was statistically significant compared with that in group B(P < 0.05).In the detection of the expression of CD62 P in the platelet membrane glycoprotein of rats after burns,it was found that the level of CD62 P of rats in group A was significantly reduced at the time points T2 and T3,and was statistically significant compared with that in group B(P < 0.05).Conclusion:After treatment with propofol,the levels of endothelin(ET-1),platelet aggregation rate(PAg R)and platelet membrane glycoprotein CD62 P in the blood of burned rats were significantly reduced,which helped to improve the hypercoagulability of blood.The mechanism of propofol inhibiting platelet aggregation may be to inhibit second phase platelet aggregation by reducing the expression of CD62 P in burn rats.
