The Effects Of Mangiferin On Human Platelet Aggregation And Secretion Of CD62P

Objective:To investigate the effects of mangiferin on human platelet aggregation and expression of CD62P in secretion after platelet activation in vitro,and study its mechanism of anti-aggregation.Method:Whole blood samples was prepared from heathy adult volunteers,incubating with the drug at 37℃,platelet activation was induced by collagen or arachidonic acid,the platelet aggregation was determined by implement's method;the percentage of CD62P-positive platelets was detected by flow cytometry.Result:100μg/ml mangiferin didn't influence the platletlet aggregation when incubated with whole blood at 5min,15min and 30min.Mangiferin could inhibit the platelet aggregation,the inhibitory effect was in a concentration-dependent manner.The amplitude of 4μg/ml,20μg/ml and 100μg/ml mangiferin induced by collagen were 17.40±4.01 ohm,15.60±4.06 ohm and 12.50±4.93 ohm,the idex induced by arachidonic acid were 28.20±7.32 ohm,21.50±7.41 ohm and 16.60±7.03 ohm,the groups of 20μg/ml and 100μg/ml mangiferin showed significance with control group(P＜0.05).The inhibitory effects induced by collagen displayed weaker than that induced by arachidonic acid at the same concentration,the IC₅₀were 140.77μg/ml and 98.61μg/ml,respectively.It could prolong the lagtime,100ug/ml mangiferin induced by arachidonic acid showed significance compared with sodium chloride(P＜0.05).But it did not influence the expression of CD62P induced by both agonists,the percentage of CD62P-positive platelets in collagen-induced platelet activation were 13.41±5.53%,15.32±8.76%and 14.29±6.60%,which were 12.29±11.48%,12.12±9.52%and 13.90±12.83%after stimulation with arachidonic acid,there was no difference between control and mangiferin-treated samples(P＞0.05).Conclusion:Mangiferin didn't influence the platelet aggregation when incubated with whole blood at different time,Mangiferin could inhibit platelet aggregation,the effect was more powerfully induced by arachidonic acid than that by collagen,but it did not affect the expression of CD62P in the course of platelet secretion.
