Liver cancer(HCC),also called hepatocellular carcinoma,is the second largest cancer causing death worldwide,threatening human health.The incidence rate and mortality rate of liver cancer in China are higher than the world average level and increase year by year.At present,surgical treatment and traditional medicine treatment can cause side effects and drug resistance.Based on the knowledge and experience of traditional Chinese medicine in treating liver cancer for a long time,what we should do is to find out the mechanism of traditional Chinese medicine on liver cancer cells on the basis of low toxicity and high efficiency,which is helpful to develop the active ingredients with anti liver cancer from traditional Chinese medicine(TCM)as soon as possible,so that more low toxicity and high efficiency anti liver cancer drugs can be applied.Therefore,nine kinds of TCM with anti hepatoma activity were selected as the research object in this paper.After soaking,boiling,concentration,constant volume,centrifugation and filtration,the traditional Chinese medicine solution was prepared.HepG2 cell line was taken as the model organism,and combined with the action mode of moxibustion therapy,the Hormesis effect mechanism of single traditional Chinese medicine solution and binary mixed traditional Chinese medicine solution on HepG2 cell was studied,As well as the effect of moxibustion alone and the combination of moxibustion and traditional Chinese medicine on HepG2 cells,and its mechanism was discussed preliminarily.The main results are as follows:(1)The preliminary mechanism of Hormesis effect on HepG2 cells was discussed from the level of cell signaling pathway.Combined with the experimental results of cytotoxic effect,cell morphology and cell cycle,it was found that TCM may change some cell signaling pathways to function:In this study,the single toxic effect of nine kinds of Chinese medicine solutions(DC,HD,MR,VA,GL,TM,SB,FO,RC)on HepG2 cells was determined.All kinds of Chinese medicine solutions(DC,HD,MR,VA,GL,TM,SB,FO,RC)had Hormesis effect on HepG2 cells.Among them,three types of HD,MR and GL with less autotoxicity have more obvious stimulating effect at low concentration,which have a greater risk of promoting HepG2 cell proliferation in a lower concentration range;The effects of TCM on HepG2 cell morphology and cell cycle were measured,and the possible mechanism of Hormesis effect of TCM on HepG2 cells was proposed.The results showed that the low concentration of TCM stimulated HepG2 cells to accelerate the cell transformation from G1 to S phase,resulting in the stimulating effect.However,as the concentration of TCM solution further increasing the accumulation of NO in cells,NO initiated the mitochondrial apoptosis pathway,inhibited cells and induced apoptosis.(2)The toxic effect of binary mixture of TCM on HepG2 cells and its joint action mode were studied:The binary combination of TCM still shows Hormesis phenomenon on HepG2 cells.According to the combined effect of three kinds of TCM liquid(HD,MR,GL)screened in the early stage,the analysis shows that the binary combination of these three kinds of TCM makes the original low concentration stimulation effect relatively reduced(the combination of HD and GL is the most obvious),and the combined toxicity is also reduced,but the maximum inhibition rate is affected.IA model was used to identify the joint toxicity of different TCM.It was found that most combinations(31groups)had low concentration synergistic or additive stimulation,medium concentration synergistic inhibition,high concentration antagonistic inhibition,only a few binary mixture of traditional Chinese medicine solutions(DC-GL,HD-GL,MR-GL,GL-SB)had low concentration antagonistic stimulation,high concentration synergistic inhibition.From the main active components of the drug itself,the mechanism of the combined action of low concentration antagonistic stimulation and high concentration synergistic inhibition on HepG2 cells was analyzed.It is suggested that the binary combination of TCM may reduce the risk of stimulating cell proliferation to some extent,but the stimulating effect still exists.Therefore,the binary combination of traditional Chinese medicine should be cautious.(3)The effects of moxibustion alone and combined with TCM on HepG2 cells were studied:With the extension of time and the increase of moxibustion times,moxibustion inhibits the growth of HepG2 cells by inhibiting the cell cycle transition from G0/G1 phase to S phase.At the same time,moxibustion can antagonize the stimulation effect of TCM on HepG2 cells when exposed to low concentration,which makes the original low concentration stimulation of TCM on cells reduce to some extent,or even disappear.The results show that the addition of moxibustion has synergistic antitumor effect on the application of TCM in the treatment of liver cancer,reducing the risk of low concentration of TCM solution on HepG2 cells to promote cell proliferation when used alone.Combined with the study of the toxic effect of moxibustion combined with TCM on HepG2 cells,and the discrimination of the mode of combined effect of TCM liquid and moxibustion on HepG2 cells,the possible mechanism of the combined effect of moxibustion and TCM liquid on HepG2 cells was explained,which provided a new idea for the research of TCM on liver cancer,and provided reference and support for moxibustion and TCM as the combined application mode of treating liver cancer.
