Rapid pathological detection has become an important application point for clinical cancer diagnosis.Microscopic observation of pathological sections is still a highly accurate detection method in cancer pathological diagnosis.However,ordinary light intensity imaging usually needs to observe the pathological sections after staining to distinguish the cancerous tissue from normal tissue.The staining of pathological sections will increase the detection time to a certain extent.Polarization imaging technology can quickly take advantage of the differences in microstructures between normal tissues and cancerous tissues,express different polarization characteristics,and then directly reflect them in their polarized images.And can directly image unstained pathological sections.Mueller matrix is a method that can fully characterize all the polarization characteristics of biological tissues.The polarization information contained in the matrix can intuitively explain the optical characteristics expressed by the internal structure of the measured tissue.With the gradual development of Mueller matrix and its analysis methods,polarization imaging technology has shown good application potential in the rapid pathological detection of cancer.Based on the advantages of polarization imaging,which is fast and non staining,and can accurately characterize the difference of biological tissue structure,a detection method of cancerous tissue based on polarization imaging is proposed in this paper.Firstly,a set of automatic imaging system for measuring the Mueller matrix of tissue scattered light was designed and built according to the double rotating wave plate method.The system uses a high-precision electronically controlled rotating stage to drive the polarization device to achieve the modulation and detection of the polarization state of light waves.Secondly,using the completed system,Mueller matrix measurement experiments were performed on unstained tissue pathological sections of breast,lung adenocarcinoma,and hepatocellular carcinoma.Finally,the Mueller matrix image data of the measured pathological slices were combined with the Mueller matrix polarization decomposition method to separate and extract polarization parameter images with clear physical meaning.The experimental results show that,compared with light intensity imaging,the corresponding Mueller matrix decomposition parameter image can clearly express the internal optical characteristics of cancerous tissue and normal tissue,and realize the distinction between the two.This provides a new reference method for rapid pathological diagnosis of breast cancer,lung adenocarcinoma and hepatocellular carcinoma.
