Backgroud:Rheumatoid arthritis(RA)is a kind of highly teratogenic autoimmune disease,which is characterized by chronic synovial inflammation,multiple joint dysfunction with bone destruction and ultimately joint function damage.More and more studies show that there is a close relationship between the cavity flora and RA.Tongue coating is the most direct expression of oral flora.The flora of tongue coating in RA patients must be different from those in healthy person,which is related to the pathogenesis of RA.However,the study on the the microecology of tongue coating in RA patients is very deficient.This study on the tongue coating microecology of RA patients is to provide a theoretical basis for the study of the cavity flora and the pathogenesis of RA.Objective:To study the relationship between disease progression and microecology of tongue coating flora in patients with rheumatoid arthritis.Methods:A total of 30 tongue coating flora swabs were collected from 10 patients in the active stage(OP,Oral Prograssive group),10 patients in the remission stage(OR,Oral Remission group)and 10 healthy women(OHC,Oral Health Control group).The total DNA extracted by the rapid oral swab genomic DNA extraction kit were amplified with 16 S rRNA and sequenced with high throughput Illumina miseq.The results were analyzed for microecology.Results:There was no statistical difference in the Alpha diversity of three groups of samples(P >0.05);three groups of samples had their own unique genera,including 7 genera in OP group,4 genera in OR group,and 10 genera in OHC group;The samples from three groups had the same composition of dominant flora,but there was significant difference in the formation of flora,mainly in p?Firmicutes,p?Actinobacteria,p?Episilonbacteraeota,p?Proteobacteria,g?Neisseria,g?Actinomycetes,g?Campylobacter,g?Porphyromonas(P <0.05).Through PCA analysis and hierarchical cluster analysis,OP group and OHC group can gather together well with significant differences.Conclusion:1.p?Bacteroides,p?Firmicutes,p?Proteobacteria,p?Fusobacteria,p?Actinobacteria,p?Patescibacteria and p?Episilonbacteraeota are the dominant bacteria in tongue coating flora of RA patients and healthy person,accounting for more than 99% of all bacteria at the level of phylum classification;2.There was no significant difference between RA patients and healthy people in the quantity and complexity of the flora in tongue coating;3.The abundance of g?Actinomycetes and g?Campylobacter increased,while that of g?Neisseria and g?Porphyromonas decreased in RA patients compared with the health control group;4.There was a significant difference between the active RA patients and the healthy group in tongue coating flora.The composition of tongue coating flora in released RA patients is changing to the healthy group.
