Objective: Osteoprotegerin(OPG),a key regulator of bone metabolism,has become a hot topic due to its role in vascular diseases and calcification.Studies have shown that OPG is associated with arteriosclerotic cerebral infarction(ACI),but its relationship with arteriosclerotic cerebral infarction(ACI)is still unclear.The relationship between OPG gene polymorphism,its plasma level and ACI,and the correlation between OPG gene polymorphism and OPG plasma level still needs further study.In this study,the relationship between plasma OPG level and OPG gene polymorphism(rs3102735)was studied in patients with large-artery artery atherosclerosis(LAA),and the role of OPG in the pathogenesis of LAA cerebral infarction was further explored.Methods: according to TOAST classification,221 patients with atherosclerotic cerebral infarction in the department of neurology of our hospital from November 2016 to November 2018 were selected,and 115 healthy subjects during the same period were selected as the control group.Plasma OPG levels of all subjects were determined by Enzyme-linked immunosorbent assay(ELISA).A Single nucleotide polymorphism(SNP)locus of rs3102735 on OPG gene was selected.The polymorphism of OPG gene was detected by Polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP).SPSS 22.0 software was used to conduct statistical analysis on the results of the study in the cerebral infarction group and the healthy control group,to analyze the level of OPG and the distribution characteristics of gene polymorphism,as well as the correlation between plasma OPG level and gene polymorphism.Results: The plasma OPG level of LAA group was higher than that of the control group(1790.71±708.39 pmol/L vs.1265.54±357.30 pmol/L,P < 0.001),indicating that the difference was statistically significant.Compared with the control group,the frequency of rs3102735 genotype and allele C in the LAA group was significantly higher(37.1% vs.24.2%,P = 0.001).20.4% vs.12.6%,P = 0.006),the difference was statistically significant.CC/CT genotypes were associated with high plasma OPG levels(2357.13±513.24pmol/L vs.1804.88±570.7 pmol/L,P = 0.003).Allele C was also associated with high plasma OPG levels(5575.94±578.77 pmol/L vs.4471.15±795.95 pmol/L,P = 0.001).Conclusion: The OPG locus rs3102735 and OPG levels may be related to the occurrence of LAA cerebral infarction.The locus rs3102735 may be involved in the occurrence of stroke by affecting OPG levels.
