Objective:To investigate the association between a 1562C→Tmutation of Matrix Metalloproteinase-9(MMP-9) gene and atherosclerosis and cerebral infarction of Shanxi.Methods:A case-control study for Shanxi Han Chinese subjects (including 112 normal control and 224 cerebral infarction) was performed. Polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP)was applied to test the MMP-9-1562 C→T polymorphism.Compare clinical variables between groups using t-test and frequencies among groups by chi-square test.All data were used to process by SPSS 17.0.Results:1.The distribution of MMP-9 genotypes in control group and Cerebral infarction group were in the Hardy-Weinberg equilibrium(P>0.05).2.C,T alleles and CC,CT genotype of MMP-9 gene promoter C-1562-T site were found in Shanxi Han Chinese subjects,but we failed to get TT genotype.3. Compared with the CC genotype, the CT genotype of MMP-9-1562 has higher level of LDL-C (P<0.05).There are no difference between other dangerous. In normal control, the CT genotype has higher level of TC and LDL-C (P<0.05). In cerebral infarction, the CT genotype has higher frequency of lipid metabolism (P<0.05).4.1n cerebral infarction, frequencies of C/C,C/T,T/Tgenetype are 83.03%,16.96%and 0,and the C, T allele frequencies are 91.52%and 8.48%. In normal control, frequencies of C/C,C/T,T/Tgenetype are 82.14%,17.86%and 0,and the C, T allele frequencies are 91.07% and 8.93%.The frequencies of genetype and allele showed no significant difference between control groups and cerebral infarction group (P>0.05).5.Taking cerebral infarction and normal control as dependent variable and genotypes of MMP-9 as independent variable,there was no difference between the CC and CT genotypes after abscise sex,age, hypertension, diabetes and hyperlipoidemia (P>0.05).6.Taking cerebral infarction and normal control as dependent variable and sex,age, hypertension, diabetes and hyperlipoidemia as independent variable,there were no difference in sex and age (P>0.05). Hypertension, diabetes and hyperlipoidemia had higher rate of cerebral infarction (P<0.05).Conclusion:1. We failed to get TT genotype of MMP-9-1562 in Shanxi Han Chinese subjects. 2.The C-1562T polymorphism of MMP-9 gene is not related to cerebral infarction in Shanxi Han Chinese subjects.3.The CT genotype of MMP-9-1562 has higher level of LDL-C (P<0.05).4.1n normal control, the CT genotype has higher level of TC and LDL-C (P<0.05).5.1n cerebral infarction, the CT genotype has higher frequency of lipid metabolism (P< 0.05)...
