Objective:Based on metabonomics research methods,observation of Sijunzi Decoction on 16 male and female mice metabolic substances in the plasma,spleen and kidney tissue and enrichment of metabolic pathways and the effect of Sijunzi Decoction in the age of mice,the mechanism of action of endogenous changes of Sijunzi Decoction intervention provide the basis for the biological basis of age-related diseases.Method:Using the natural aging model,ICR mice were fed normally to the age of3 months and 16 months.The 3-month-old mice were selected as the blank group(3M group),and the 16-month-old naturally aging mice were randomly divided into the model group(16M group)and the drug administration group(SJZ group),with 20 mice in each group(10 females and 10 males).Mice in the drug group were given iv junzi decoction by intragastric administration(the dose was 9.75g/kg·d,and the volume was 13ml/kg·d).Mice in the blank group and the model group were given normal saline by intragastric administration.After continuous gavage for 30 days,plasma,spleen and kidney tissue samples were prepared and metabolomic analysis was performed using uplc-q-tof-ms.Results:1.After sijunzi decoction intervention,metabolites in blood,spleen and kidney tissues of mice significantly changes in metabolic profile,the three samples of PCA analysis and processing,display blank(3 m),model group(16 m),the drug group(SJZ)data to distinguish between good and drug metabolites to the expression of blank group callback.2.Regulation of sijunzi decoction on age-related marker differences in plasma,spleen and kidney tissue samples:1)plasma samples(16 m)in the model group compared with the blank group(3 m),select biomarkers related to age,including 32 in male mice,there are 36 in female mice,identified the biological markers associated withseven metabolic pathways,including male and female mice iconic metabolites concentration respectively in four metabolic pathways;In the sijunzi decoction group,19 metabolites in male mice and 16 metabolites in female mice could be recalled.The recalled metabolites were related to 8 metabolic pathways,among which the marker metabolites in male and female mice were respectively enriched in 4 metabolic pathways.2)will the spleen tissue samples(16 m)in the model group compared with the blank group(3 m),screening identified biomarkers associated with age,with 59 in male mice,with 69 in female mice,identified the biomarkers related to article 21 of metabolic pathways,including male and female mice iconic metabolites concentration respectively in article 17 and article 7 in the metabolic pathway,there are three common metabolic pathways for the male female mice;The sijunzi decoction group could recall 53 metabolites in male mice and 19 metabolites in female mice.The metabolites of the callback were related to 19 metabolic pathways,among which the marker metabolites in male and female mice were enriched in 16 and 3 metabolic pathways respectively.3)renal tissue samples(16 m)in the model group compared with the blank group(3 m),screening identified biomarkers related to increased age male mice,70,67 in female mice,identified the biological markers associated with article 25 metabolic pathways,including male and female mice iconic metabolites concentration in article 20 and 17,respectively,metabolic pathway,there are 12 pathway for male female mice common metabolic pathways;In the sijunzi decoction group,54 metabolites in male mice and 39 metabolites in female mice could be recalled.The recalled metabolites were related to 27 metabolic pathways,among which the marker metabolites in male and female mice were respectively enriched in 17 and 10 metabolic pathways.3.Regulation of sijunzi decoction on age-related marker differences in different genders of 16-month-old mice:1)in male mice,sijunzi decoction affects the metabolism of plasma,spleen and kidney by regulating docosahexaenoic acid and l-carnitine metabolism.In plasma and kidney tissue samples,5 of the same metabolites were affected.In the samples of kidney and spleen,10 of the same metabolites were affected.Four of the same metabolites were affected in plasma and spleen tissue samples.2)in female rats,sijunzi decoction can affect the metabolism of plasma,spleen and kidney by regulating the metabolism of13s-hydroxyoctadecarboxylic acid.In the metabolism of metabolites in plasma and kidney tissue samples,4 common substances were affected.Three of the same metabolites were affected in plasma and spleen tissue samples.Seven of the same metabolites were affected in both kidney and spleen tissue samples.4.Regulation of sijunzi decoction on age-related marker differences in different samples of 16-month-old mice:1)in the plasma samples,sijunzi decoction intervened the physiological state of male and female mice by regulating 4 substances such as estrone,docosahexaenoic acid and l-dihydrowhey acid.2)in the spleen tissue samples,sijunzi decoction intervened the physiological state of male and female mice by regulating 9 substances such as 16-a-hydroxyprogestenolone,sulfonylcholine glycine and l-glutamic acid.3)in kidney tissue samples,sijunzi decoction intervened the physiological state of male and female mice by regulating 19 substances such as l-phenylalanine,l-tryptophan,FAD,flavin mononucleotide and creatine.Conclusion:1.(16 m)mice model group compared with the blank group(3 m)in mice,plasma,spleen and kidney tissue samples metabolic marker are different degrees of change,mice under natural state of growth and development during the metabolism have obvious changes,the metabolite markers mainlyconcentrated in the amino acid metabolism and lipid metabolism in related pathways.2.Sijunzi decoction for 16 months in plasma,spleen and kidney tissue samples of age related metabolites have certain callback function,the callback substances were involved in the plasma,spleen and kidney tissue samples of article 8,19,27 age-related metabolic pathways,focused primarily on amino acid metabolism and lipid metabolism related way.3.The metabolic pathways mainly regulated by sijunzi decoction are similar to those affected by aging changes in mice,indicating that sijunzi decoction can regulate the changes of metabolism of all tissues caused by aging.4.Sijunzi decoction had a regulatory effect on age-related marker differences in 16-month-old mice of different genders and tissues.
