The Experimental Study On Effect Of IG, IL-18and5-HT Of Aging Model Mice By Four Tonic Prescriptions

Objective:By studying the subacute aging model mice’s content of IgA, IgG of blood serum and the content of IL-18,5-HT of skin, the subject explored the aging model mice’s immunologic function among four tonic prescriptions as Sijunzi Decoction, Siwu Decoction, Liuweidihuang Decoction and Jinkuishenqi Decoction, to further elucidate the modern immunological characteristics of deficiency syndrome.Methods:Two months of age Kunming mice were randomly divided into six groups, which included four drug groups, as Sijunzi Decoction, Siwu Decoction, Liuweidihuang Decoction and Jinkuishenqi Decoction, D-Gal group and blank control group, there were16mice in each group, male and female are both8mice. Four drug groups and D-Gal group were alternately taken by abdominal injection and hypodermic injection to duplicate subacute aging mice model. Blank control group were injected normal saline in the same way. Four drug groups were treated with the method of oral administration two hours later, and the rest of the two groups were filled normal saline. In the experiment of the42th day, every rat was taken eyes to gather blood and separated serum, to detect the content of IgA and IgG of serum with the method of immunonephelometry. Killed the mouse and used their10%homogenate of skin to detect the content of IL-18and5-HT with the method of immunoenzymatic techniques.Results:(1) Compared with the blank control group, global D-Gal groups can reduce IgA level obviously(P<0.01). Compared with D-Gal group, the IgA content of the four prescriptions provided groups were increased significantly (P<0.01).Compared four tonic prescriptions global effect, Liuweidihuang Decoction>Sijunzi Decoction>Siwu Decoction>Jinkuishenqi Decoction. The effect of IgA caused by Siwu Decoction is that male mice are superior to female mice. The effect of IgA caused by Jinkuishenqi Decoction is that female mice are superior to male mice.(2) Compared with the blank control group, global D-Gal groups can reduce IgG level obviously (P<0.01). Compared with D-Gal group, Liuweidihuang Decoction, Jinkuishenqi Decoction and Siwu Decoction groups can increase IgG level obviously (P<0.01), Sijunzi Decoction can increase IgG level (P<0.05).Compared four tonic prescriptions global effect, Liuweidihuang Decoction>Jinkuishenqi Decoction>Siwu Decoction>Sijunzi Decoction. The effect of IgG caused by Liuweidihuang Decoction is that female mice are superior to male mice. The effect of IgG caused by Jinkuishenqi Decoction and Sijunzi Decoction is that male mice are superior to female mice.(3) Compared with the blank control group, global D-Gal groups can rise IL-18level (P<0.05). Compared with D-Gal group, the IL-18content of Siwu Decoction reduced (P<0.05), Sijunzi Decoction, Jinkuishenqi Decoction and Liuweidihuang Decoction had downtrend, but the change was not very obvious. Compared four tonic prescriptions global effect, Siwu Decoction>Sijunzi Decoction>Jinkuishenqi Decoction>Liuweidihuang Decoction.(4) Compared with the blank control group, global D-Gal groups can raise5-HT level obviously (P<0.01). Compared with D-Gal group, the5-HT content of Liuweidihuang Decoction and Sijunzi Decoction reduced (P<0.05), Siwu Decoction and Jinkuishenqi Decoction had downtrend, but the change was not very obvious. Compared four tonic prescriptions global effect, Liuweidihuang Decoction>Sijunzi Decoction>Jinkuishenqi Decoction>Siwu Decoction. The effect of5-HT caused by Liuweidihuang Decoction and Sijunzi Decoction is that male mice are superior to female mice.Conclusion:All of four tonic prescriptions can increase the content of IgA and IgG in serum of Sub acute Aging Model Mice, and reduce the level of IL-18and5-HT in skin, but the extent of adjusting effect is different, to prove the four tonic prescriptions have immunologic function. It was confirmed important significance of Immunology by nourishing process of Qi-Tonics, Blood-Tonics, Yin-Tonics and Yang-Tonics and tonic prescriptions, in which, compared four tonic prescriptions global effect on the content of IgA, Liuweidihuang Decoction>Sijunzi Decoction>Siwu Decoction>Jinkuishenqi Decoction. Compared four tonic prescriptions global effect on the content of IgG, Liuweidihuang Decoction>Jinkuishenqi Decoction>Siwu Decoction>Sijunzi Decoction. Compared four tonic prescriptions global effect on the content of IL-18, Siwu Decoction>Sijunzi Decoction>Jinkuishenqi Decoction>Liuweidihuang Decoction. Compared four tonic prescriptions global effect on the content of5-HT, Liuweidihuang Decoction>Sijunzi Decoction>Jinkuishenqi Decoction>Siwu Decoction. Changes in the level of IgG and5-HT caused by aging, and changes in the level of IgA, IgG and5-HT caused by taking four tonic prescriptions, are associated with the mice’s gender.The subject, to further explore the modern immunological characteristics of deficiency syndrome, made a theoretical guidance for further study on the immunosenescence mechanism of classic tonic prescription regulating action.
