Skin aging is one of the most common aging phenomena in the human body,which is generally divided into chronologic aging and photoaging.Time aging is charactered by tiny wrinkle,skin chalasis,arescent and rough skin,while photoaging is charactered by out of elast- icity,big wrinkle and leather-skin generally companied with coloring material.Traditional Chinese medicine is a safe and effective way against skin aging, and the theories in TCM consider that Spleen-stomach system and nutrient qi-defense qi system have great relationship with skin aging.In the literature view,this dissertation discuss the relationship between Spleen- stomach system and nutrient qi-defense qi system and skin aging,which can give theorical support to the methods of strengthening Spleen-stomach system and supplementing Ying-wei against skin aging.1 Literature ResearchThis part of the paper maily reviewed the following content: the origin and development of the Chinese medical theory about skin aging,the effects of the spleen-stomach against aging and skin aging,the relationship between Ying-wei,skin aging and aging.The developments of the Chinese medical theory about skin aging and aging were traced back to the same origin.They germinated before the chunqiu dynasty,formed in the Han and Tang Dynasties,enriched in the Song,Yuan,Ming and Qing Dynasties,and developed in the present day.《Shang Han Za Bing Lun》written by Zhang zhong jing paid attention to supplement the Ying-wei and strengthen the spleen-stomach.The relationship between the Ying-wei and skin aging were discussed clearly.The spleen-stomach govern transforming and transporting food and water.They are vie- wed as the sources for the production and transformation of qi and blood,as well as the acquired foundation of the body.The spleen-stomach which located in the middle jiao are the center of qi action.the ascending,descending,exiting and entering of qi rely on the function of the spleen-stomach.Their functions are easily injured by diet regulation,person's taste,life style and emotions.That lead to deficienly of qi and blood,lack of the Ying-wei,result in skin aging.Dysfunction of the spleen-stomach can easily lead to problems of phlegm and blood stagnantion,thus skin aging would exacerbated.The Ying-wei are produced in the middle jiao,conveged in the skin.They have the foll- owing functions:nutrishing and proctecting the skin,against exogenous pathogens,roling sen- ses of the skin,adjusting the excretion of sweat,etc.These functions are related with the skin physiology and pathology.The Ying normally circulates in the vessels while the Wei outside the blood vessels.They communicate with each other by meridianns,points,collaterals,sweat pore and interstitial space to keep the balance of the Yin and Yang.The circulation of Ying-wei are rhythmical.The dysfunction of the Ying-wei is the basical pathological factor which lead to skin aging.2 Experiment research2.1 ObjectiveDiscuss the deep mechanism of supplementing Ying-wei against skin aging according to observing anti-oxydation of supplementing Ying-wei and the influence of supplementing Ying -wei to collagen of aging skin.2.2 Method120 virgin mice were randomly divided into 6 groups.One group is the control group, conventional feeding.The other 5 groups were given subcutaneouly injections daily with the dose of D-galactose(120mg/kg·d-1) in the skin.Of the 5 groups,one group is the model group treated with daily subcutaneouly injection of saline,conventional feeding,light.One group sets the control group treated with daily intragastric administration with vitamin E(50mg/ kg·d-1).The other three groups were treated dividedly with 8g/kg·d-1,4g/kg·d-1 and 2g/kg·d-1 decoction of supplementing Ying-wei for consecutive six weeks.Finally take the blood samp- les from the abdominal aortic,separating the blood serum and using the kit to determinate the activity of superoxide dismutase(SOD) and the contents of malondialdehyde(MDA) and hyaluronic acid.Then precipitate the erythrocyte,washing,using kit to determinate the activity of Na+,K+-ATPase.Use the depilatory skin of the back to produce 10% homogenates,using the kit to determinate the activity of superoxide dismutase(SOD),the contents of malondial- dehyde(MDA) and hyaluronic acid and the concentration protein.Take an appropriate amount of depilatory skin, using the kit to determinate its hydroxyproline content.Take the neck skin into 4% paraformaldehyde-fixed,paraffin-embedded,HE staining and masson staining,and microscopic observation of histopathological changes.In addition,sections in 5μm,using imm- uneohistochemical methods and image analysis system to evaluate the expression of trans- forming growth factor beta 1(TGF-1),connective tissue growth factor(CTGF),basic fibroblast growth factor(bFGF),Matrix metalloproteinase(MMP),MMP-1 and MMP-9,inhibitors of matrix metalloproteinases(TIMPs)TIMP-1,organizational plasminogen activator(t-PA) and the organization of plasminogen activator inhibitor-1(PAI-1) in aging skin.2.3 ResultsAfter six weeks,of the mice treated with subcutaneouly injection of D-galactose (120mg /kg·d-1),in the Model group the contents of MDA in blood serum increased signifycantly and the activity of SOD reduced,the results was statistically significant compared with other groups.Taking in supplementing Ying-wei can lower the contents of MDA,and the high-dose supplementing Ying-wei group was better than the positive group(P<0.05),low-dose group (P<0.001) and the control group (P<0.05).Supplementing Ying-wei can improve blood SOD activity.The blood SOD activity in low-dose group and middle-dose group was equal to the control group while was statistically significant compared with the model group(P<0.05).The blood SOD activity in high-dose group was statistically significant compared with the positive group(P<0.05).After the model, the MDA contents in skin increased significantly,significantly higher than that of other groups.And the SOD activity decreased.Taking in supplementing Ying-wei can lower the MDA contents and high-dose group is better than the positive group(P<0.05), low-dose group(P<0.05) and the control group(P<0.01).Taking in supplementing Ying-wei can improve blood SOD activity.The blood SOD activity in low-dose group and middle-dose group was equal to the control group while was statistically significant compared with the model group(P<0.05).The blood SOD activity in high-dose group was statistically significant compared with the positive group(P<0.05)and was higher than the control group.After the model, the protein contents in skin decreased,and the result was statistically significant compared with other groups(P<0.05,P<0.001 or P<0.001).After taking in suppl- ementing Ying-wei,the protein contents increased significantly in the high,middle and low dose group and were higher than the control group,which the results in high-dose group were statistically significant(P<0.05) compared with the positive group and the low-dose group. After D-galactose subcutaneouly injection,the skin hydroxyproline contents decreased and the result was statistically significant compared with other groups(P<0.05 or P<0.001).Taking in supplementing Ying-wei can significantly increase the skin hydroxyproline level.The skin hydroxyproline level in low-dose group was equal to the control group.The skin hydro- xyproline level in middle and high dose group were higher than the control and positive group and the results were statistically significant compared with the positive group(P<0.001 and P<0.05)and the control group(P<0.001).The skin hydroxyproline level in high-dose group were higher than the low-dose group(P<0.05).Na+,K+-ATPase activity reduced and there is a significant difference(P<0.001)comp- ared with other groups.Taking in supplementing Ying-wei can improve Na+,K+-ATPase activity in erythrocyte.Na+,K+-ATPase activity in erythrocyte in low-dose group and middle- dose group was equal to the control group.Na+,K+-ATPase activity in high-dose group was statistically significant compared with the control group(P<0.05)and the low-dose group(P< 0.001).Hyaluronic acid in the blood serum by analysis of variance,the variance of each group were different,and with rank and testing,there is no statistical significance(P>0.05).Compared with the control mice skin structure,the skin structure of mice in model group is identified clearly,and epidermal and dermal boundaries is still clear.But the epiderm has no rules and dermis is significantly thinner,deeper staining,less cell layers.Subcut-aneouly fat is rich.Hair follicles and sebaceous gland reduced.After taking in supplementing Ying-wei and positive group,there is the structural integrity in the skin of mice,and epidermal and dermal line is clear.Epiderm layer is in rule.Dermis layer is thicker than control group and cell layers increased.there is less subcutaneouly fat. After the model,the bFGF expression level was significantly lower,and compared with control group there was statistically significant(p<0.05).After taking supplementing Ying-wei, bFGF levels increase significantly,and compared with the control group, model group, positive group,and low-dose group there were significantly different(p<0.05,p<0.001,p<0.05 and p<0.05,respectively).The low-dose group and positive group made the low bFGF level rebound to the normally level.The content of protein expression in high-dose group increased compared with the control group and there was statistically significant difference(p<0.05).After the model,TGF-β1 expression level was significantly decreased.Compared with control group there was statistically significant(p<0.05).After taking in supplementing Ying- wei,TGF-β1 level of low,middle and high group improved significantly,and compared with model group there was statistically significant.Positive group can increase the reduced TGF-β1 level,and compared with the model group there was statistically significant(P<0.05).After the model,CTGF expression level significantly improved, and compared with control group and the supplementing Ying-wei group there was statistically significant(p<0.05).After taking supplementing Ying-wei,CTGF expression level is equal to that of the control group, and there was no significant difference among in high,middle and low dose groups (p>0.05).After taking in VE,CTGF expression level in hair follicles and other places signi- ficantly increased,and compared with the other groups there was a significant difference.After the model,MMP-1 expression levels increased significantly, and compared with the control group there was statistically significant(p<0.05).After taking in supplementing Ying-wei, MMP-1 levels in high,middle and low dose group increased slightly and compared with the model group there was no significant difference(p>0.05).After taking in VE,inc- reased MMP-1 level reduced,and compared with the model group and high-dose group there was statistically significant(P<0.05),and also was lower than supplementing Ying-wei grou- ps.After the model,MMP-9 expression level was significantly lower compared with the control group,and there was a significant difference(P<0.05).After taking in supplementing Ying-wei,MMP-9 expression levels increased significantly,and compared with the model group there was statistically significant(p<0.05).And there was no significant difference among in high,middle and low dose groups(p>0.05).After taking in VE, MMP-9 expression level significantly increased,and compared with the model groups there was a significant difference(p<0.05).After the model,the expression of TIMP-1 levels significantly decreased,and compared with the other groups there were statistically significant(p<0.01).The TIMP-1 levels in the supplementing Ying-wei groups increased,but there was no significant difference among in high,middle and low dose groups(p>0.05).After taking in VE,increased TIMP-1 level redu- ced,and compared with the model group there was statistically significant(P<0.01).After the model, TIMP-2 expression levels increased,and compared with the group there was statis- tically significant(P<0.05).After taking supplementing Ying-wei,TIMP-2 expression level is equal to that of the control group,and there was no significant difference among in hig- h,middle and low dose groups(p>0.05).After taking in VE,TIMP-2 expression level sign- ificantly decreased.After the model,tPA expression level increased significantly,and compared with the other groups there are statistically significant(p<0.01).The tPA levels in the supplementing Ying-wei groups increased,and compared with the model group there are statistically significant(p<0.01),but there was no significant difference in high,middle and low dose groups(p>0.05).After the model,the PAI-1 expression levels changed slightly,and compared with the control group,supplementing Ying-weigroups there was not statistically significant (p>0.05).After taking in supplementing Ying-wei,tPA expression level is equal to that of the control group,and there was no significant difference among in high,middle and low dose groups(p>0.05).After taking in VE,CTGF expression level in hair follicles and other places significantly increased,and compared with the other groups there was a significant difference. 2.4 ConclusionSupplementing Ying-wei can oppose oxidization and increase the skin protein synthesis and collagen content so that it can delay skin aging.
