Objective: This research investigates the correlations of lung injury to the risk factors and to abnormal expression of phosphate and tension homology deleted on chromsome ten(PTEN)in pneumoconiosis.This will provide new ideas and experimental data for the early prevention,the early detection and early diagnosis of pneumoconiosis..Methods: 130 patients diagnosed with pneumoconiosis(65 patients with stage I pneumoconiosis and 65 patients with stage II)in Hunan Chest Hospital and 50 cases of healthy controls were selected as study members.Lung tissue sections and computer tomography(CT)were employed to investigate the histological and radiographic features of pneumoconiosis.By investigating and analyzing the history of pneumoconiosis,the correlation between risk factors(age,education level,smoking status,tuberculosis complications,dust exposure time)and lung injury was explored.Western blot and ELISA were used to detect and compare the expression level of PTEN in lung tissues and peripheral blood of patients with pneumoconiosis at different stages,and Pearson correlation coefficient was used to expound the correlation between the expression level of PTEN in lung tissues and peripheral blood.Finally,PTEN expression in peripheral blood of pneumoconiosis patients with different stages and different lung injuries was detected,and the relationship between PTEN expression in peripheral blood of pneumoconiosis patients and lung injury was analyzed.Results:1.Lung tissue sections showed the destruction of alveolar structure in the pneumoconiosis group,which was replaced by silicon nodules.The nodules presented hyaline degeneration,and the surrounding interstitial collagen fibrosis was accompanied by dust deposition.The results of Computed Tomography(CT)showed that the lungs were diffusely distributed in different sizes and densities with nodular shadow,accompanied by patchy cord strip shadow and slightly thickened pleura.The results suggested that the lung injury of pneumoconiosis patients was obvious.2.Risk factors for the study subjects(1)Comparing the general demographic indicators,there were significant differences in the composition of educational level between the pneumoconiosis group and the normal control group.The majority of pneumoconiosis patients were low-educated people.The results showed that the age and educational level of stage II pneumoconiosis group were lower than that of stage I pneumoconiosis group.(2)the pneumoconiosis group and the normal control group,There were significant differences in the distribution of smoking status,smoking index and tuberculosis between the two groups.(3).The results showed that there were significant differences in smoking status,smoking index,concomitant tuberculosis and dust exposure time between the two groups.3.Correlation analysis of high risk factors and pneumoconiosis injury in patients with pneumoconiosis:(1)There was statistical difference in lung injury among different education levels,lung injury is related to education level;(2)There was statistical difference in lung injury between smoking group and non-smoking group,lung injury is related to educational level;(3)There was statistical difference between tuberculosis group and nontuberculosis group,pulmonary injury is related to tuberculosis;(4)The degree of lung injury was different in different dust exposure time groups,moderate positive correlation was significantly shown between the exposure time and the degree of pneumoconiosis injury,and the correlation was moderate.4.Analysis of PTEN expression in pneumoconiosis patients with different stages:(1)the expression of PTEN in biopsy lung tissues of 15 cases of stage I pneumoconiosis group was significantly higher than that of 15 cases of stage II pneumoconiosis group.(2)PTEN expression in peripheral blood of stage I and II pneumoconiosis group was significantly lower than that of the normal control group,and the difference was statistically significant.PTEN expression in peripheral blood of stage II pneumoconiosis group was significantly lower than that of stage I pneumoconiosis group,and the difference was statistically significant.(3)the Pearson correlation coefficient of PTEN expression in peripheral blood and lung tissues of pneumoconiosis patients was 0.919,suggesting that PTEN expression in peripheral blood and lung tissues of pneumoconiosis patients was positively correlated and consistent.(4)correlation analysis between PTEN expression level in peripheral blood and pneumoconiosis lung injury: in stage I pneumoconiosis patients,the mean PTEN expression level in peripheral blood of patients with mild injury was 196.54±9.57 pg/ml,and the mean PTEN expression level in patients with moderate injury was 154.26±9.7 pg/ml,and the difference between the two groups was significant.In patients with stage II pneumoconiosis,the mean PTEN expression level in peripheral blood of patients with mild injury was 152.16±7.51 pg/ml,the mean PTEN expression level in peripheral blood of patients with moderate injury was 113.73±7.17 pg/ml,the mean PTEN expression level in peripheral blood of patients with severe injury was 80.8±8.76 pg/ml.There was a significant difference between the three groups,the results suggested that the less the expression of PTEN in peripheral blood,the more severe the degree of lung injury.Conclusion: 1.The risk factors of pneumoconiosis(age,education level,smoking status,tuberculosis complication,dust exposure time)are correlated with pneumoconiosis lung injury.2.PTEN expression in pneumoconiosis tissues and peripheral blood has a good consistency,showing a positive correlation,and PTEN expression in peripheral blood is negatively correlated with pneumoconiosis lung injury.
