ObjectiveHashimoto’s thyroiditis(HT)is an immunological thyroid disease that significantly affects the female reproductive process.Galectin-1 was found to be one of the most important proteins that may be involved in a woman’s pregnancy.Therefore,the expression of galectin-1 in patients with HT may be influenced to some extent by their own diseases.In this paper,we studied the expression of galectin-1,and the expression of tumor necrosis factor-α(TNF-α),and interleukin-17a(IL-17 a)in the endometrial estrous cycle of rats with HT.The objective is to clarify the mechanism of HT affecting female reproductive process.Methods40 healthy NOD female mice of 8-9 weeks in age were randomly divided into two groups(by random number method): normal control group(CON group)and Hashimoto thyroiditis group(HT group),20 in each group.A mouse model of Hashimoto’s thyroiditis was established by subcutaneous injection of porcine thyroglobulin(p Tg)into the caudal root of mice.The estrus cycle of mice was determined by Rayleigh staining.Respectively to compare two groups of mice serum thyroglobulin antibodies(Tg Ab),thyroid peroxidase(TPOAb),triiodothyronine(T3),four iodine glycine(T4)and thyroid-stimulating hormone(TSH)level,the level of estrogen periods of the endometrial thickness,estrogen galectin-1 m RNA expression in every period,the estrogen from one period to the average optical density value of galectin-1 protein(AOD),uterine tissue homogenate tumor necrosis factor-α(TNF-α),interleukin-17 aResults1.The serum Tg Ab and TPOAb levels in the observation group were(207.95±14.75)IU/mol/L and(98.44±5.10)IU/mol/L,respectively,which were higher than those in the control group(173.11±8.26)IU/mol/L and(84.56±4.77)IU/mol/L(all P < 0.05).However,serum levels of T3,T4 and TSH in the HT group were(1.44±0.30)nmol/L,(35.38±7.49)nmol/L and(355.62±2.31)μmol/L,which were not significant compared with those in the CON group(1.42±0.28)nmol/L,(35.02±7.50)nmol/L and(354.11±3.05)μmol/L(all P > 0.05).2.The endometrial thickness of mice in the HT group was(377.53±36.20)μm,which was thicker than(341.32±20.15)μm in the CON group(P < 0.05).The endometrial thickness of mice in the HT group was(354.28±18.57)μm,(367.32±14.18)μm,(248.82±13.48)μm,respectively,compared with that in the CON group(352.31±17.65)μm,(364.57±14.23)μm,(250.31±12.67)μm,respectively(all P > 0.05).3.The expression levels of galectin-1 m RNA in the HT group were(0.67±0.05),(0.54±0.02),(0.52±0.03),(0.50±0.03),(1.64±0.10),(1.28±0.06),(1.23±0.06),and(1.19±0.06),respectively,which were lower than those in the CON group(P < 0.05).In the HT group,the AOD of galectin-1 protein was(0.1657±0.0029)and(0.0814±0.0021),respectively,which were lower than that of the CON group(0.2018±0.0036)and(0.1397±0.0020)(P < 0.05),while the AOD of the HT group was(0.0831±0.0025)and(0.0428±0.0027),respectively.There was no significant difference(P > 0.05)between the two groups(0.0829±0.0023)and(0.0433±0.0026).4.The levels of TNF-α and IL-17 a in the tissue homogenate of mice in the HT group were(0.92±0.11)pg/ml and(0.36±0.02)pg/ml,respectively,which were lower than those in CON group(1.57±0.25)pg/ml and(0.68±0.15)pg/ml,(all P < 0.05).(IL-17 a).Conclusion: The endometrium of Hashimoto’s thyroiditis mice with normal thyroid function showed immune response,low expression of galectin-1,and endometrial thickening in the estrus phase.Galectin-1 was expressed in the endometrial estrous period of Hashimoto’s thyroiditis mice,which was lower than that of the CON group,which was related to the local cytokine changes in the uterus of Hashimoto’s thyroiditis mice to some extent.
