Objective Hashimoto’s thyroiditis(HT)can causes female infertility,and estrogen receptors play a major role in a woman’s pregnancy.In this study,we tested the estrous cycle by administering NOD female mouse porcine thyroglobulin(p Tg).The estrous cycle was detected to observe whether the expression of estrogen receptor alpha in endometrium of HT mice was affected during the estrous,and the local cytokine level of uterus was detected to understand the local immunity of HT in uterus,and to explore the influence of HT mice on pregnancy.Methods Thirty healthy NOD mice were selected and standardized for 8-9 weeks old,fe male,and clean grade.After feeding for a week,the mice were randomly div ided into two groups,called Hashimoto thyroiditis group(HT group),normal cont rol group(CON group).Pour out 100 μl of PBS solution and add 25 μg of porc ine thyroglobulin p Tg to it.CFA and p Tg solution were mixed 1:1 in a mortar,and ground to complete emulsification under ice bath conditions.mice tail root was injected with 0.1ml emulsifier.the same dose of CFA was given to the nor mal control group.After 14 days,CFA was replaced with IFA in the HT group,a nd by the same method,IFA was given the equal doses in the control group.Th e 49-day molding period.Thyroid specimens of mice were collected for HE stai ning and X-ray observation.Serum samples of mice were collected to detect thyroid peroxidase antibody(TPOAb),T3,T4,thyroglobulin antibody(Tg Ab)and TSH.Collect vaginal exfoliated cells for Swiss staining to detect estrous cycle in mice.Immunohistochemical staining was used to detect the expression and distribution of estrogen receptor alpha in mouse endometrium.and the uterine estrogen rece ptor alpha,cytokines IL-6 and IL-10 were detected by PCR.Cytometric Bead Arra y(CBA)method was used to detect the levels of interleukin-6 andinterleukin-10 in the two groups.Result1.Mice thyroid gross specimen results: all HT mice thyroid increased compared with the normal control group.The color of the thyroid tissue is reddish,and the texture is tough and has a nodular sensation.Result of HE thyroid staining : Compared with the normal control group,the lymphocytes infiltration and the destruction of some thyroid follicles were observed in the thyroid gland of the Hashimoto group.2.Thyroid hormone and Tg Ab and TPOAb levels of mice in two groups:Compared with CON group,there were no difference in TSH,T3 and T4 between HT group and the normal group.Compared with CON group,Tg Ab of HT group were increased obviously(p<0.001).TPOAb of HT group were elevated(p<0.05).3.Immunohistochemistry results of both groups:The expression of ERα immunpositive cells in eutopic endometrium of CON group and HT group.ERα was expressed in luminal epithelium,glandular epitheliumand stroma of the endometrium.Positive substances are mainly present in the nucleus,and cytoplasm is also expressed.The level of ERα in the endometrium of the CON group was higher than that in the HT group(P<0.05).4.PCR results: The ERα m RNA levels in the estrus of the CON group were higher than those in HT group(P<0.001);the IL-6 m RNA level was lower in the HT group than in the CON group(P<0.001),and IL-10 m RNA level was lower in the HT group than in the CON group(P<0.001).5.The results of Wright’s staining in CON and HT groups showed that there were nuclear epithelial cells and a few keratinized epithelial cells in the visual field as pre-estrus;a large number of large keratinized acellular cells with a small number of nucleated epithelial cells as estrus phase;white blood cells,keratinized epithelial cells and nuclear epithelial cells all had post-estrus phase;a large number of white blood cells and a small number of epithelial cells existed as estrus phase.6.The levelof IL-6 in the CON group were higher than those in the HT group(P<0.05).The level of IL-10 in the CON group were higher than those in the HT group(P<0.001).Conclusion HT causes a decrease in the expression of ERα in the endometrium of mice,and the levels of IL-6 and IL-10 in the uterus were changed.The results of this experiment may have some reference significance for the study of HT affecting pregnancy.
