Purpose Detection of KIAA1199 gene expression in pancreatic cancer tissues and analysis of its correlation with clinicopathological factors and prognosis.Methods Collect 60 pathologically diagnosed pancreatic cancer tissues and 20 of them to obtain pancreatic tissues adjacent to the cancer.Perform immunohistochemical staining experiments to detect the expression of KIAA1199 gene in paraffin sections.And analyze its relationship with related clinicopathological factors and survival time of pancreatic cancer.At the same time,Western Blot and q RT-PCR experiments were used to detect the expression of KIAA1199 in 10 fresh frozen pancreatic cancer specimens and corresponding adjacent tissues.Results Immunohistochemical results showed that the positive expression rate of KIAA1199 gene in pancreatic cancer was 70%,which was significantly higher than the positive expression rate of 25% in adjacent tissues,and the difference was statistically significant(P < 0.05).The expression of KIAA1199 gene was related to the pathological differentiation of pancreatic cancer(P = 0.01),TNM stage(P = 0.047),nerve infiltration(P = 0.016),and lymph node metastasis(P < 0.05),and the differences were statistically significant(P < 0.05).There was no significant correlation with the patient's gender(P = 0.515),age(P = 0.206),and tumor location(P = 0.726).Kaplan-Meier survival analysis showed that OS and DFS of KIAA1199 positive expression was significantly shorter than that of negative expression,with a significant difference(P < 0.05).WB and q RT-PCR experiments confirmed that the expression of KIAA1199 in pancreatic cancer tissues was higher than that in adjacent tissues,and the difference was statistically significant(P = 0.0001,P < 0.0001).Conclusion KIAA1199 gene is highly expressed in pancreatic cancer tissues and is expected to be an effective predictor of tumor progression and prognosis in patients with pancreatic cancer.
