Objective To investigate the expression of human kallikrein 11(KLK11)in pancreatic cancer tissue(PCT)and non-cancerous normal pancreatic tissue(Non-PCT)and its clinical significance.Methods The expression of KLK11 in PCT and Non-PCT paired with 75 pancreatic cancer patients was detected by immunohistochemical(IHC)method,and the relationship between KLK11 expression and clinical pathological parameters and prognosis was also analyzed.The expression of KLK11 in 10 matched pairs of PCT and Non-PCT was tested by Western blot and real-time fluorescence quantitative PCR(qRT-PCR).Results IHC showed that KLK11 was mainly expressed in the cytoplasm of PCT ductal cells.In the PCT group,the positive expression rate of KLK11 reached 77.33%(58/75),while in the Non-PCT group the rate was 20%(15/75),the difference between the two groups was statistically significant(P<0.001).The expression of KLK11 has a significant correlation with lymph node metastasis(P<0.001)and TNM staging(P<0.001).Western blot and qRT-PCR showed that the expression and mRNA content of KLK11 in PCT group were significantly higher than those in Non-PCT group,these two different statistics showed the meaning(P<0.01,P<0.001).Through the analysis of Kaplan-Meier,the result showed that the survival time of KLK11 high expression(average DFS: 7.2 months,mean OS: 14.3 months)was significantly shorter than that of KLK11 with low expression time(average DFS: 17.0 months,mean OS: 22.8 months).The difference was statistically significant(P < 0.01).The analysis of COX regression model showed that KLK11 expression is an independent factor in the prognosis of pancreatic cancer.Statistically significant(P<0.01).Conclusion The expression of KLK11 is closely related to lymph node metastasis and TNM staging of pancreatic cancer,which can be used as an independent factor to determine the prognosis of pancreatic cancer.The detection of KLK11 has a certain value in the occurance and development of pancreatic cancer and the evaluation of its prognosis. |
