| Objective To observe the effect of mesencephalic astrocyte-derived neurotrophic factor(MANF)on experimental autoimmune uveitis(EAU)in rats.Methods Thirty Lewis rats were randomly divided into three groups: normal group,EAU experimental model group(control group)and EAU experimental model treatment group(experimental group).rats in the experimental group and the control group were immunized successively with the combination of complete freund’s adjuvant and pertussis toxin with the photoreceptor intercellular vitamin a binding protein(IRBP)to make an eau model.the rats in the normal group did not do any treatment(three groups were fed in the same feeding environment).after that,the experimental group was given manf via vitreous cavity on the 3rd,7th,11 th day after immunization.the control group was given phosphate solution(PBS)as positive control in vitreous cavity on the same day.the normal group was not treated.On the0 th,7th and 14 th days after immunization,the present arthritis in rats was observed under slit lamp and the degree of inflammation was scored by Caspase.rats were sacrificed at the peak of inflammation,and hematoxylin-eosin staining was used to compare the retinal inflammatory response and tissue morphology of the three groups of rats.The expression of interleukin-17(IL-17)and interferon-gamma(INF-gamma)m RNAN was detected by real-time fluorescence quantitative PCR(RT-q PCR)Results After successful modeling,three groups of rats were observed by slit lamp,and the control group began to develop inflammation on the 3rd day after immunization,and the iris vasodilation was seen under the slit lamp.On the seventh day after immunization,mild inflammation was observed under slit lamp.The 14 th day is the peak period of inflammation,the pupil membrane is closed under the slit lamp,the pathological histological results show that the inflammatory cells are obviously infiltrated,the retinal structure is seriously damaged and the neovascularization appears.The rats in the experimental group also developed inflammation on the third day after immunization.On the seventh day after immunization,only moderate hyperemia was seen in the iris and no turbidity was seen in the anterior chamber.However,on the 14 th day after immunization,histopathology and clinical manifestations suggested that the ocular inflammation in the experimental group was significantly improved compared with the control group,the anterior chamber was only mildly turbid,and the histopathological retinal structure was intact,with only a small number of neovascularizations.RT-q PCR results suggested that the expression levels of inflammatory factors IL-17 and INF-γ;in the experimental group were significantly lower than those in the control group(p <0.05).Conclusion MANF vitreous cavity administration can reduce the inflammation of EAU in rats,and the decrease of the expressions of IL-17 and INF-γ may be a part of the effect,which may provide new ideas for the treatment of uveitis. |
