Background Experimental autoimmune uveitis(EAU)is a common animal model of uveitis.The present study was designed to explore the role and mechanism of NK cells in the pathogenesis of EAU.Objective To explore the location of NK cells in the retinal tissue of the EAU at different stages,and test the expression of chemokines in different stages of EAU.Methods 61 Lewis rats were randomly divided into two groups: normal control group and EAU experiment model group.Rats in normal control group are given with saline combined with complete Freund's adjuvant,and 400 ng pertussis toxin successively.Rats in EAU group are given the interphotoreceptor retinoid binding protein(IRBP)combined with complete Freund's adjuvant,and 400 ng pertussis toxin successively.From day 1 to day 21 after immunization,we use the slit lamp microscope to observe anterior segment.The score of ocular inflammation was evaluated according to Caspi's method,and evaluating scores of retina inflammation at 3,6,9,12,16,21 day after immunizing.HE staining was employed to detect the infiltration of inflammatory cells and the destruction of tissue structure in Lewis rats.Immunofluorescence method was used to detect the distribution and infiltration of NK cells in retina.RT-PCR method was used to detect the expression of NK cell chemokine m RNA in eyeball.Results EAU group appeared inflammatory performance and retinal tissue began to swell in the antigen immunization 6 days.The peaks of inflammation and inflammatorycells infiltration were observed in 12 days.We can see following manifestations: serious damage in the organizational structure,the both scores of anterior segment inflammation and inflammation of the retina are 4.Inflammation and inflammatory cells disappear and retinal structure gradually recovery at 21 day after immunization.NK cells were seen at 6 day after immunizing.The number of NK cells significantly elevated at 9day,and was significantly less at 12 day than 9 days after immunizing;NK cells disappeared in 16 days.The expression of CXCL10 has statistically increased at 9 day compared with other time points after immunizing(F=16.997,P<0.01),and CXCL12 were no difference at different time points(F=2.851,P>0.05).Conclusions NK cells play an important role in the early stage of EAU,and CXCL10 is an important chemokine of NK cells in EAU.
