Objectives:In order to elucidate the pathogenesis of xuanwei lung cancer,and the potential biomarkers related to the pathogenesis of xuanwei lung cancer could be selected as the prognostic factors of xuanwei lung cancer by proteomics,so as to improve the diagnosis,treatment and prognosis assessment of xuanwei lung cancer patients.Methods:Proteins in lung cancer tissues and paired normal tissues were extracted by trypsin enzymolysis,proteome sequencing was performed by TMT labeling,and target proteins were screened by bioinformatics software including protein annotation,subcellular localization,differential protein screening,GO enrichment,and protein interaction network.Results:In all samples,there were more than 100 differentially expressed proteins in cancer tissues/adjacent normal tissues,of which 382 were differentially expressed proteins in lung cancer tissues and normal lung tissues,189 were up-regulated and 193 were down-regulated.GO analysis suggested that most of the differentially expressed proteins have the function of transport and are the main components of cell membrane and play a binding role.KEGG signaling pathway enrichment analysis revealed that nitrogen metabolism and Hematopoietic cell lineage pathway were significantly enriched.Conclusions:We identified at least 5,991 proteins in each sample,among which 382 were differentially expressed in cancer tissues and normal lung tissues by bioinformatics analysis.The enrichment analysis of gene ontology indicated that most of the differentially expressed proteins were proteins with transport function,among which,the low expressed proteins were mainly related to the nitrogen metabolism pathway,while the high expressed proteins were mainly related to the Hematopoietic cell lineage pathway.Comparing with TCGA and other databases,there was significant differences in the transcription level and protein level of CA4 gene in the nitrogen metabolism pathway,and the expression of CA4 gene decreased with the tumor stages.Meanwhile,CA4 gene could also serve as a potential prognostic marker of lung cancer.
