There are two parts in this study:(1)Evaluation of the feasibility of acute carotid thrombosis model induced by FeC13 in rabbits by Carotid Doppler ultrasonography(CDU);(2)To establish the model of ischemic stroke induced by carotid thrombosis.Part ? Evaluation of the feasibility of acute carotid thrombosis model induced by FeC13 in rabbits by Carotid Doppler ultrasonographyObjective:To explore the feasibility of vascular ultrasound in evaluating the rabbit model of carotid thrombosis induced by FeCl3 infiltration,and to establish a simple,rapid and consistent with the mechanism of clinical spontaneous thrombosis.Method:1.Group of model rabbits:acute carotid thrombosis model was established by using the chemical oxidation of FeC13 to induce carotid intimal injury.Nine New Zealand white rabbits were randomly divided into three groups(A,B,C),three in each group.70%FeCl3 was used to infiltrate the left common carotid artery,10 mins in group A,20 mins in group B and 30 mins in group C,respectively.Before infiltration,D-dimer was detected in three groups of rabbits.2.Anesthesia and ultrasonic detection:100mg ketamine and 10mg diazepam were diluted to 8ml with 0.9%normal saline,and the first dose was lml/kg.After intramuscular injection,anesthesia was started.The diameter of left common carotid artery and peak systolic velocity were measured with 14L high-frequency linear array probe.After that,the left common carotid artery was dissected,and the corresponding time(10mins,20mins,30mins)of FeC13 filter paper was infiltrated.After the infiltration,it was sutured layer by layer.Then,the peak systolic velocity of the common carotid artery in the experimental section was measured by body surface ultrasound,and the proportion of thrombus in the lumen was evaluated.3.Pathological examination:after the end of the experiment,the common carotid artery of the experimental section was taken out,fixed with 4%formaldehyde,embedded in paraffin,sectioned,and detected by HE staining.4.Statistical methods:SPSS 17.0 software was used for statistical analysis.The mean± standard deviation(x±s)of the measurement data indicates that the analysis of variance is used for the comparison between multiple groups,and LSD test is used for the comparison between the two groups.The difference was statistically significant when p<0.05.Result:1.Baseline data of three groups of model rabbits before modeling:Before 70%FeC13 infiltration,the diameter of the common carotid artery in the three groups were 1.730±0.106mm,1.670 ± 0.183mm,1.670 ± 0.216mm(p>0.05),the peak systolic velocity was 77.461±9.123cm/s,76.345±6.551cm/s,78.617±7.471 cm/s,(p>0.05),and the blood D-dimer index was 0.133±0.016mg/L,0.121±0.018 mg/L,0.127±0.019mg/L(p>0.05),respectively.2.Ultrasonic examination results of three groups of model rabbits after modeling:there was no significant thrombosis in the experimental carotid artery of group A,and there was no significant difference in the peak systolic velocity(P>0.05);non obstructive thrombosis was found in group B,the proportion of thrombus in lumen was 58.10 ±9.83%,and the peak systolic velocity was significantly faster than that before modeling(P<0.05);obstructive thrombosis and vascular occlusion were found in group C.3.Pathological examination results:there was no thrombus in group A,but mixed thrombus in group B and C.Conclusion:The 70%FeC13-induced rabbit carotid artery thrombosis model is reliable and fast,and its pathological structure is closer to the histological and morphological characteristics of clinical spontaneous thrombosis,which lays the experimental foundation for the future thrombolytic treatment.Part ? To establish the model of ischemic stroke induced by carotid thrombosis.Objective:To explore the success rate of establishing ischemic stroke model with FeCl3 at different infiltration time.Method:1.Group of model rabbits:24 New Zealand white rabbits were randomly divided into 3 groups(group A,group B,group C),8 in each group.70%FeCl3 was used to infiltrate the left common carotid artery,10 mins in group A,20 mins in group B and 30 mins in group C respectively.2.Nerve defect score:24 hours later,the nerve function defect score(Purdy score)was randomly given to the three groups of model rabbits.3.MRI:at the end of Purdy score,the brain of three groups of model rabbits was examined by MRI,and the examination sequence was DWI,T1WI,T2WI-FLAIR.4.Pathological examination:after euthanasia,the brain tissue was taken out,fixed with formaldehyde,embedded in paraffin,sectioned and stained with he to evaluate the success rate of ischemic stroke model in each group.5.Statistical analysis:SPSS 17.0 software was used for statistical analysis.The mean±standard deviation(x±s)of the measurement data indicates that the variance analysis is used for the comparison between multiple groups,and LSD test is used for the comparison between the two groups;the frequency(%)is used for the calculation data,and Fisher's accurate probability method is used for the comparison.The difference was statistically significant when p<0.05.Result:1.Purdy score:Purdy score of group A,B and C were 1.700±0.949,6.000 ± 1.155 and 8.400 ± 2.171,respectively.Purdy score of group C was higher than that of group A and B(P<0.05);2.MRI results:no abnormal signal was found in group A;no abnormal signal was found in group B DWI,T1WI and T2WI-FLAIR;no abnormal signal was found in group C DWI,T1WI and T2WI-FLAIR;3.Pathological examination results:there was no abnormality in HE staining of group A,and infarct was found in HE staining of group B and C.4.Modeling success rate:the modeling success rates of group A,group B and group C were 0,100%and 62.5%respectively.Conclusion:FeC13-induced acute carotid thrombosis in rabbits can be used to prepare focal cerebral ischemia model,infiltrate carotid artery for 20 minutes,and the success rate of modeling is higher.
