Purpose:We study the effects of IVF-ET(in vitro fertilization-embryo transfer)on the metabolism of glucose and lipids in offspring relating regulatory mechanisms.Methods:We establish the mouse model of IVF-ET offspring to study the effect of IVF-ET on the glucose and lipid metabolism of offspring comparing with the normal-conceived offspring.At the age of 12 weeks,male mice were selected from each group for experiments.We measure the body weight and insulin secretion of every selected mice;after 12 h of fasting,performing the insulin tolerance test and glucose tolerance test.After the mice were sacrificed,liver tissues were collected for liver glycogen PAS staining and fatty oil red staining;ELISA kit methods were used to quantify liver glycogen and triglyceride content;RT-PCR was used to determine the expression of genes related to glucose and lipid metabolism;measuring the pAKT protein expression by using the Western blot.Results:The IVF-ET group's body weight was increased compared with normal-conceived mice,and no change in insulin secretion.IVF-ET mice have impaired glucose tolerance;reducing the liver glycogen compared with the normal-conceived group,increasing the liver triglyceride compared with the normal-conceived group;expression of genes regulating liver glycogen synthesis lowering than the normal-conceived group;increasing the expression of liver glucose metabolism and fat synthesis genes compared with the normal-conceived group;the expression of liver pAKT protein lowering than that of the normal-conceived group.Conclusion:IVF-ET mice have abnormal expression of liver glycogen synthesis genes and proteins,which indirectly leads to abnormal expression of liver glucose and fat metabolism genes,causing of abnormal glucose and lipid metabolism in mice. |
