Objective:To explore the expression and clinical significance of chemokine-like factor-like MARVEL transmembrane domain-containing family member 6(CMTM6)in breast cancer,and to analyze the relationship between CMTM6 and the clinicopathological characteristics and prognosis of breast cancer patients,and to evaluate the diagnostic value of CMTM6 in breast cancer,so as to provide a basis for diagnosis and evaluation of breast cancer and a new theoretical basis for immunotherapy of breast cancer.Methods:The expression of CMTM6 was detected by immunohistochemistry in 136 breast cancer tissue chips,including 42 matched cancer and paracancerous tissues.?2 test was used to compare the expression difference of CMTM6 in breast cancer and adjacent tissues,and to analyse the relationship between the expression level of CMTM6 in breast cancer and the clinicopathological characteristics of patients.The relationship between the expression level of CMTM6 and the survival period of breast cancer patients was analyzed by Kaplan-Meier and Log-rank,and Cox model was used to evaluate the relationship between different indicators and the prognosis of patients.At the same time,serum samples and clinical data of 49 breast cancer patients,39 fibroma patients and 26 healthy people were collected and the concentration of CMTM6 was detected by enzyme-linked immunosorbent assay.Wilcoxon's rank sum test was used to compare the CMTM6 concentrations of breast cancer patients,fibroma patients and physical examination healthy people in the serum.The relationship between the CMTM6 concentration of breast cancer patients in the serum and their clinicopathological characteristics were analyzed.The concentrations of CA153 and CEA in serum samples were measured by electrochemiluminescence,and TK1 in serum samples by enzyme immunoblotting,and ROC curves were drawn to evaluate the diagnostic value of serum CMTM6,CA153,CEA and TK1 in breast cancer.Results:Immunohistochemical staining showed that CMTM6 was widely expressed in breast cancer tissues,and the high expression rate of CMTM6 in breast cancer tissues was significantly higher than that in adjacent tissues(P<0.01).In addition,the expression of CMTM6 was correlated with pathological type and HER2 positive of breast cancer(P<0.05).Kaplan-Meier survival analysis showed that the survival time of CMTM6 high expression group was significantly shorter than that of CMTM6 low expression group(P=0.038).Multivariate Cox proportional risk model showed that pathological type(HR=10.374,95%CI:3.529?30.497,P<0.001),TNM stage(HR=4.599,95%CI:1.784?11.856,P=0.002),triple-negative breast cancer(HR=3.370,95%CI:1.055?10.761,P=0.04)and CMTM6 high expression(HR=0.195,95%CI:0.073?0.518,P=0.001)were independent risk factors for prognosis of breast cancer patients.The results of serum samples showed that the concentration of CMTM6 in breast cancer patients and fibroma patients was significantly higher than that in healthy people(P<0.01).In addition,the CMTM6 concentration in the serum was correlated with pathological type(P=0.023),lymph node metastasis(P=0.048)and TNM stage(P=0.037).Analysis of the diagnostic value of serum CMTM6,CA153,CEA and TK1 for breast cancer showed that CMTM6 alone had the highest sensitivity(93.88%)and lowest specificity(43.75%).Combined diagnosis of CMTM6,CA153,CEA and TK1 for breast cancer has the highest AUC(0.830).CMTM6 combined with CA153,CEA and TK1 can improve the specificity and accuracy of breast cancer diagnosis.Conclusions:CMTM6 is widely expressed in breast cancer,and its expression is significantly up-regulated compared with the adjacent tissues.The expression of CMTM6 is significantly up-regulated in invasive ductal cancer and HER2 positive patients.The high expression of CMTM6 is significantly related to the prognosis and survival of breast cancer patients.The level of CMTM6 in the serum of breast cancer patients was significantly higher than that of the healthy people,and it was related to the pathological type,lymph node metastasis and TNM stage of breast cancer.CMTM6 combined with CA153,CEA and TK1 can improve the specificity and accuracy of breast cancer diagnosis,and has high diagnostic value.
