Objective:We aimed to evaluate the expression levels of CKLF-like MARVEL transmembrane domain-containing 6(CMTM6)in lung cancer,to explore the correlation between it and the clinicopathologic features and prognosis and survival of patients,and provide new strategies and theoretical basis for the assessment,monitoring and immunotherapy of patients with lung cancerMethods:A total of 86 paraffin-embedded tumor specimens were selected after HE staining reading,pathological diagnosis confirmed as lung adenocarcinoma,and complete follow-up data were collected.86 lung tissues over 2cm from the edge of corresponding lung cancer tissues were taken as paracancerous tissue,and all patients received no treatment before surgery.The expression of CMTM6 in lung cancer and adjacent tissues was determined by tissue microarray and immunohistochemical staining.Meanwhile,52 patients diagnosed with lung cancer and 32 healthy people were selected,and the expression levels of CMTM6 and PD-L1 before and after surgery were detected by ELISA,and the correlation between each group was analyzed.Results:Immunohistochemical results showed that the CMTM6 was widely expressed in 86 lung cancer tissues,compared with the corresponding adjacent tissues,30%showed upregulation and 70%showed no difference.The up-regulated proportion of CMTM6 in the high-level group of clinical stage was significantly higher than that in the low-level group(?2=4.269,P=0.033),and the up-regulated proportion of CMTM6 in the distant metastasis group of lung cancer was significantly higher than that in the non-distant metastasis group(?2=6.234,P=0.046).In the clinical staging group comparison,the survival rate of the CMTM6 up-regulated group was significantly lower than that of the CMTM6 expression unchanged group(P=0.001)in the stage ? patients with lung cancer.Kaplan-meier survival analysis showed that the difference of CMTM6 expression was statistically significant associated with patient survival(P=0.014),and the survival rate of patients in the up-regulation group was significantly lower than that in the unchanged group.Multivariate analysis using Cox regression model showed that the expression of CMTM6 was an independent prognostic factor for postoperative survival of lung cancer patients.ELISA experiments had shown that CMTM6 preoperative surgery in lung cancer,the physical examination the expression level of serum difference between three groups was statistically significant(P<0.05),Spearman correlation analysis showed a significant correlation between CMTM6 and PD-L1(r=0.623,P<0.01),serum CMTM6 expression in tumor diameter of 3.5 cm or more group were significantly higher than that of tumor diameter<3.5 cm group(P=0.014),the high expression rate of serum CMTM6 in patients aged 65 years or older was significantly higher than that in patients<65years old.(P=0.001).Conclusions:CMTM6 was widely expressed in lung cancer tissues and adjacent tissues,and the expression of CMTM6 was significantly up-regulated in patients with high clinical staging of lung cancer and distant metastasis.The expression state of CMTM6 was closely related to postoperative survival of patients with lung cancer.The expression level of CMTM6 in peripheral blood was significantly correlated with the level of PD-L1,the serum level of CMTM6 in lung cancer patients was significantly higher than that in healthy people,and the serum level of CMTM6 was significantly correlated with the patient's age and tumor size.CMTM6 is an independent prognostic factor in the development of lung cancer...
