Objective: The purpose of this study was to observe the effect of huoshenjiangzhuo decoction on urinary protein excretion rate,biochemical indexes,fibrogenic factor expression and pathological damage status of kidney tissue in adriamycin nephropathy rats.This study provides a theoretical basis for delaying renal fibrosis in rats with adriamycin nephropathy.To investigate huoshenjiangzhuo decoction in the treatment of chronic kidney disease(CKD)mechanism,It provides some thoughts for the direction of clinical medication of huoshenjiangzhuo decoction.Methods: A pathological model of adriamycin nephropathy was established by injecting adriamycin into the tail vein of male Wistar rats.It was divided into four groups:huoshenjiangzhuo Decoction group(treatment group),nephritis rehabilitation tablet group(control group),model group and blank group.The general state,urinary microalbumin /creatinine,24-hour urinary protein,serum creatinine and urea nitrogen of rats were observed.Kidney sections were stained with masu to observe the pathological changes of kidney tissues in rats with adriamycin nephropathy.The pathological tissue structure of kidney was observed under light microscope.Immunohistochemistry was used to observe the expression of p-JAK2,p-STAT3,NF-?B and PGSK-3?.Results: 1.After treatment,compared to the model group,the serum creatinine,urea nitrogen,all urinary albumin / urinary creatinine and 24-hour urine protein in the treatment group compared with the model group,a statistically significant difference(P<0.05).2.Immunohistochemical results showed that compared with the model group,the deliver of pJAK2,p-STAT3,NF-?B,and PGSK-3? in the treatment group and the control group decreased in glomerular endothelial cells,mesangial cells and epithelial cells,with statistically significant differences(p<0.05).3.Pathomorphological observation of kidney tissues: no obvious pathological changes were observed in glomerulus,renal tubules and interstitium in the blank group,with normal arrangement and distribution.The number of fibrous cells in the model increased significantly,the number of collagen fibers increased,the fiber bundles were thickened and disarranged,and the fiber gaps were narrowed or disappeared.With the progress of experiment time,the number of aidan collagen fibers in interstitium increased gradually,indicating the progressive aggravation of fibrosis.Partialtubules fibrosis,focal interstitial fibrosis,renal interstitial collagen fiber increased in the treatment group and control group.Conclusion: 1.Huoshenjiangzhuo decoction can reduce the 24-hour urinary protein ration and urinary microalbumin/urinary creatinine ratio in adriamycin nephropathy rats,and reduce the excretion of urinary protein;2.The expression of p-JAK2,p-STAT3,NF-?B,PGSK-3? in renal tissues of rats with adriamycin nephropathy was down-regulated by Huoshenjiangzhuo decoction;3.Huoshenjiangzhuo decoction can improve the process of renal fibrosis in adriamycin nephropathy rats.
