Objective:To study the expression of heat shock factor 1(HSF1)and microvessel density(MVD)in patients with pancreatic cancer and to elucidate the relevance between HSF1 and angiogenesis,clinicopathologic factors and prognosis.Methods:A total of 196 paraffin-embedded pancreatic cancer and paracancerous tissue samples were randomly collected from patients diagnosed with pancreatic cancer from January 2007 to September 2013 and undergoing pancreatectomy and 6 fresh pancreatic cancer tissues surgically removed in 2017 were collected during autopsy performed by an expert.Collect personal information and clinical characteristics of patients,mainly including gender(male/female),age(>60 years old/<60 years old),CEA(with 5ng/m as the threshold),CA-199(with 100ng/ml as the threshold)Value),tumor diameter(with 2cm as the cut-off point),lymph node metastasis,TNM staging(according to the TNM staging system revised by the International Anti-Cancer Alliance(UICC)in 2002),the location of the tumor(head/pancreatic body and Pancreatic tail),differentiation(can be divided into high,middle and low differentiation),whether there is vascular infiltration,whether there is a tumor residue.The expression of HSF1 protein was detected by immunohistochemical staining and western blot;and the expression of CD34 protein was measured to represent MVD.Statistical analysis of HSF1 expression,MVD and collected clinical variables;correlation analysis of HSF1 and MVD;statistical analysis of HSF1 expression results,MVD and tumor recurrence and survivalResults:1.According to the results of QRT-PCR and Western bolt,compared with normal pancreas and adjacent tissues,HSF1 mRNA expression was significantly increased in pancreatic cancer tissues.2.According to the results of immunohistochemistry,compared with normal pancreas and adjacent tissues,HSF1 protein is significantly increased in pancreatic cancer tissues.3.According to the results of immunohistochemistry,CD34 was significantly overexpressed in pancreatic cancer tissue,suggesting that MVD was significantly increased in pancreatic cancer.4.High expression of HSF1 and high MVD are significantly associated with disease-free survival.The overall survival and disease-specific survival of patients with high HSF1 expression and high MVD were significantly reduced.5.High expression of HSF1 and CA-199 levels,whether lymph node metastasis,TNM staging,tumor cell differentiation,vascular invasion,and tumor residual correlation P values are 0.002,0.005,0.003,0.011 0.035,0.023,P<0.05,suggesting significant correlation;while the P values of gender,age,CEA,tumor maximum diameter,and tumor location were 0.172,0.832,0.255,0.812,0.195,and P>0.05,respectively,suggesting no significant difference.6.Increased MVD and CA-199 levels(P=0.001,P<0.01),lymph node metastasis(P=0.035,P<0.01),TNM stage(P<0.001),differentiation(P<0.001),vascular infiltration(P=0.007,P<0.01),residual tumors(P=0.024,P<0.05)were significantly related,and were related to gender(P=0.338,P>0.05),age(P=0.99,P>0.05),CEA(P=0.13,P>0.05),tumor diameter(P=0.271,P>0.05),tumor location(P=0.71,P>0.05)showed no significant difference.7.Multivariate analysis showed that high expression of HSF1 is an independent prognostic factor for overall disease specificity and disease-free survival.Conclusions:HSF1 is highly expressed in pancreatic cancer tissues,which is related to the prognosis of patients.The higher the expression,the worse the prognosis.It may be used as a new therapeutic target for pancreatic cancer and a biological indicator of prognosis.
