Objective:To prove the important role of vasoactive intestinal peptide(VIP)in the lateral geniculate body(LGBd)in visual development and explore the therapeutic effect of VIP in amblyopia.Methods:3-week-old domestic cats were randomly divided into control group(n=10)and monocular deprivation group(n=20).The right eyes of the deprivation group were covered with black opaque blindfold.The pattern visual evoked potential(PVEP)was used to detect the formation of monocular amblyopia in the deprivation group.The left lateral geniculate body was isolated from 5 kittens in each group.The kittens in the remaining control group were reared as the normal control group,and the kittens in the residual deprivation group were divided into VIP intervention group(n=5),Sefsol(caprylic acid monoglyceride,VIP solvent)intervention group(n=5)and amblyopia non-intervention group(n=5).After 3 weeks,the latency and amplitude of P100 wave,the number of positive cells and the average optical density of VIP immunohistochemistry and VIP-mRNA in situ hybridization in the left lateral geniculate body of each group were compared.Results:After 3 weeks of monocular occlusion(6 weeks old),there were significant differences in the results of P100 wave,VIP immunohistochemistry and in situ hybridization between the normal group and the deprivation group.After 3 weeks of VIP intervention,the results of P100 wave in VIP intervention group were better than those in Sefsol intervention group and amblyopia non-intervention group(P<0.05),but still different from those in normal control group(P<0.05).The number of positive cells in VIP immunohistochemistry was increased(P<0.05),and the average optical density of positive cells was increased(P>0.05),which was still lower than that in normal control group(P<0.05).The number of positive cells in VIP-mRNA in situ hybridization was increased(P<0.05),and the average optical density of positive cells was increased(P<0.05),which was still lower than that in normal control group(P<0.05).Conclusion:VIP in the lateral geniculate body plays an important role in visual development.Exogenous administration of VIP can improve the function of neurons in the lateral geniculate body of kittens and has a certain therapeutic effect on amblyopia.
