Experimental Study On The Treatment By Chinese Herbs Of Constipation-predominant Irritable Bowel Syndrome SD Rats

Objective:Constipation type irritable bowel syndrome(IBS-C)is a common functional bowel disease in clinic,western medicine treatment of this disease is mostly limited to a single target,Chinese medicine can play a holistic therapeutic role,which Chinese medicine on the basis of shugan has achieved good clinical effect in treating the disease.We investigated the effects of Bupleurum Shugan Powder and Jichuan decoction on the gastrointestinal hormones 5-HT,SS and VIP in IBS-C model rats to explore its possible mechanism.Methods:SD rats were randomly divided into normal group(8)and IBS-C model group(48).Normal group has normal feeding without special treatment.In the model group,ice water gavage was used to limit drinking water combined with chronic restraint stress.The model-making successful SD rats were randomly divided into model control group,compound polyet-hylene glycol electrolyte powder(PEG-EP)group,Delixin group,low dose group,medium dose group and high dose group of traditional Chinese medicine.After the treatment was completed,the defecation-related indexes,the relevant indexes of the open field experiment,the intestinal pressure threshold,the percentage of sugar preference,the small intestine propulsion rate,the determination of 5-HT SSand VIP content in the serum were determined in each group of rats.Results:Compared with the model group,Bupleurum Shugan Powder and Jichuan decoction significantly increased the fecal number,fecal water content,Bristol score,open field test score,visceral sensitivity score,sugar preference rate and intestinal propulsion rate in IBS-C model rats(P<0.01 or P<0.05),the contents of 5-HT and SS in serum decreased(P<0.05),and the contents of VIP in serum increased(P<0.05).Compared with PEG-EP group and Deanxit group,Chinese medicine high dose group rat serum 5-HT and SS were significantly decreased(P<0.05).Significant increase in VIP level(P<0.05).Conclusion:1.Ice water gavage combined with limited drinking water,chronic restraint stress model making method is simple,the effect is stable,can simulate the liver depression qi stagnation combined with yang deficiency type IBS-C rats;2.Compared with PEG-EP group and Deanxit group,the serum 5-HT and SS of Chinese medicine medium dose group,high dose group were significantly decreased,and the level of VIP in the serum of rats was significantly increased,indicating that traditional Chinese medicine was better than western medicine,and there was a quantitative effect relationship between the Chinese medicine treatment effect,in a certain range there is a tendency to increase the effect with the increase of dose,the high dose of traditional Chinese medicine may be the best dose;3.The increase of reward interest in sugar water,the increase of horizontal activity score and the decrease of intestinal sensitivity after treatment in the Deanxit group,which indicated that the antidepressant Deanxit was effective in the treatment of IBS-C.
